Blood transcriptomic data from PRAM-D cohort

ABSTRACT: Pregnant women suffering from depression show alterations in different biological systems, but the exact underlying mechanisms are still not clear. We used microarrays to investigate the transcriptomic modulation in conditions such as depression and risk for developing depression during pregnancy.

Project description:DNA methylation profiles were generating using Illumina HM450 microarrays in a prospective sample blood from the prenatal period of pregnant mood disorder patients who would and would not develop depression post partum. We recruited 54 pregnant women with a history of either Major Depression or Bipolar Disorder (I, II or NOS) and prospectively followed them during pregnancy and after delivery in order to identify genetic and clinical characteristics that precede the development of a postpartum depressive episode. Blood samples profiled were collected at varying time points during pregnancy.

Project description:To explore the influence of choline intake and pregnancy status on gene expression, we employed whole genome microarray expression profiling to identify genes that were differentially expressed between two choline intake groups and between pregnant and non-pregnant women. Healthy third trimester (gestational week 26-29) pregnant women and non-pregnant women were randomized to a 12-week choline controlled feeding study. The participants consumed either 480 (n=6 pregnant and n=5 non-pregnant) or 930 (n=6 pregnant and n=5 non-pregnant) mg choline/d. Fasting peripheral blood leukocyte samples were collected at week 0 and week 12 to extract RNA and perform the arrays. Healthy third trimester (gestational week 26-29) pregnant women and non-pregnant women were randomized to a 12-week choline controlled feeding study. The participants consumed either 480 (n=6 pregnant and n=5 non-pregnant) or 930 (n=6 pregnant and n=5 non-pregnant) mg choline/d for 12 weeks. Fasting (10-h) peripheral blood leukocyte gene expression were measured at week 0 and week 12.

We then performed differential expression analyses to assess the miRNA profile diffrences according to the presence of pregnancy complications or not (i.e. Gestational diabetes mellitus, Gestational hypertension or preeclampsia vs. normal pregnancies).

Loss of endometrial plasticity in recurrent pregnancy loss (RNA-Seq)

Project description:In order to try and identify characteristics of gene expression in the endometrium of women suffering infertility or recurrenty miscarriage, we performed RNAseq on endometrial pipelle biopsies from 20 women. The endometrial transcriptome in the mid-luteal phase of the cycle (window of implantation) is highly divergent in women suffering infertility or miscarriages. 20 mid-luteal endometrial biopsies were analysed from infertile women and patients suffering recurrent pregnancy loss.

Analysis of plasma proteome of pregnant women with polycystic ovary syndrome (PCOS

Project description:Polycystic ovary syndrome (PCOS) is the most common endocrinological disorder of fertile-aged women. PCOS has been associated with adverse pregnancy outcomes and abnormalities of the placenta. By taking a quantitative label-free quantitative proteomics approach we set out to investigate if changes in the plasma proteome of pregnant women with PCOS could elucidate the mechanisms behind the pathologies observed in PCOS pregnancies. We have performed label-free quantitative proteomics on plasma samples from pregnant women with PCOS at term (n=14) and plasma samples from pregnant control women (n = 23) matched for age, gestational length and BMI. The samples are derived from BASIC pregnancy cohort from Uppsala, Sweden. A total of 169 proteins with two or more unique peptides were identified.

EV-microRNA Signatures in Pregnant Women with Idiopathic Recurrent Pregnancy Loss: Deciphering MicroRNAome Pathway Networks at Feto-Maternal Interface

Project description:Despite extensive research in the past decade, the exact pathogenesis of recurrent pregnancy loss (RPL) remains unknown. At the time of pregnancy, human placenta releases microRNAs (miRNAs) enclosed in extracellular vesicles (EVs), which enter into maternal circulation and play an important role at feto-maternal interface to sustain a successful pregnancy. Aberrant expression of these miRNAs often results in adverse pregnancy complications. Therefore, studying the expression of these EV-miRNAs in maternal circulation could provide insights into the pathogenesis of RPL. miRNA sequencing revealed 66 (44 known and 22 novel) significantly DE miRNAs between RPL and healthy pregnant women.

Normal early pregnancy: a transient state of epigenetic change favoring hypomethylation

Project description:The objective of this study was to analyze genome-wide differential methylation patterns in maternal leukocyte DNA in early pregnant and non-pregnant states. This is an age- and body mass index-matched case-control study comparing the methylation patterns of 27,578 cytosine-guanine (CpG) sites in 14,495 genes in maternal leukocyte DNA in early pregnancy (n=14), in the same women postpartum (n=14), and in nulligravid women (n=14) on a BeadChip platform. Transient widespread hypomethylation was found in early pregnancy as compared with the non-pregnant states. Methylation of nine genes was significantly different in early pregnancy compared to both postpartum and nulligravid states (< 10% False Discovery Rate). Early pregnancy may be characterized by widespread hypomethylation compared to non-pregnant states; there is no apparent permanent methylation imprint after a normal-term gestation. Nine potential candidate genes were identified as differentially methylated in early pregnancy and may play a role in the maternal adaptation to pregnancy.
