Project description:This study aims to investigate the transcriptomic differences between wild-type Arabidopsis thaliana Col-0 and the odr1-2 mutant in two experimental groups: imbibed seeds (under normal hydration conditions) and seeds subjected to 12-hour mannitol treatment (a common osmotic stress mimic).Using RNA sequencing to compare global gene expression profiles, we seek to identify differentially expressed genes (DEGs) associated with osmotic stress responses, seed imbibition, and the potential regulatory role of the ODR1 gene. Our analysis will clarify how the ODR1 mutation alters transcriptional dynamics under both normal and osmotic stress conditions, thereby providing insights into the molecular mechanisms underlying stress tolerance and seed physiology in Arabidopsis.

Gene expression between dry and imbibed seeds in vitamin C defective (vtc) mutants and wild-type Arabidopsis thaliana

Project description:In the dry state, orthodox seeds have little antioxidant capacity, relying on reduced glutathione and tocochromanols for antioxidant defences rather than ascorbate or ascorbate peroxidase (APX). Ascorbate is synthesised de novo within a few hours of imbibition resulting in rapid increases in the ascorbate pool in the developing embryo. The knock-on effects of enhanced oxidation on embryo development before and after imbibition remain poorly characterised. A T-DNA insertional mutant line (SAIL_769_H05) line was used in these studies together with vtc2-1 mutants that were originally identified in an ethyl-methanesulfonate (EMS) mutagenesis screen. Expression levels of dry seeds and imbibed seeds of the genotypes Col0, vtc2 (EMS) and vtc2.5 (SAIL_769_H05) were quantified using Illumina RNA-seq technology

Gene expression in dormant, germinating and thermoinhibited Arabidopsis seeds

Project description:To establish the basis for understanding molecular mechanism of seed germination response to temperature, we analyzed transcriptomes in freshly harvested dormant and dry stored after-ripened seeds. The after-ripened seeds started to show visible germination from 36h after the start of imbibition, and almost all the seeds germinated after 3 days. The freshly harvested seeds stayed dormant by imbibition at 26°C, and germination of the after-ripened seeds was almost completely inhibited at 34°C. Total RNA was prepared from 0 (dry), 6 and 24h imbibed seeds to find regulatory genes of seed dormancy and germination.
