Melatonin analog-agomelatine alleviates hypoxia-induced pulmonary arterial hypertension by activating mitophagy via the SIRT1/FoxO1/ULK1 signaling pathway

ABSTRACT: To explore the protective effect and molecular mechanism of agomelatine in hypoxic pulmonary arterial hypertension, we used Su5416 combined with hypoxia-induced rat PAH model and hypoxia-induced PASMC model. We verified agomelatine's therapeutic effect both in vivo and in vitro. Then, we performed RNA sequencing on control, SuHx, and SuHx+AGM groups. Pathway enrichment analysis revealed increased mitophagy, confirmed by WB, laser confocal microscopy, and electron microscopy. Finally, we found that agomelatine alleviates hypoxic pulmonary arterial hypertension by regulating mitophagy through the SIRT1/FOXO1/ULK1 pathway.
