Genome-wide binding profiles of Sp8 in differentiation mouse embryonic stem cells

ABSTRACT: We have determined the genome-wide binding profile of the transcription factor Sp8. We found that Flag-Sp8 prefentially bound to promoters associatd with several growth factor signalling pathways, but most prominently with the Wnt/beta-catenin pathway to selectively promote mesoderm differentiation over neural. These data implicate Sp8 as new regulator of Wnt/beta-catenin target expression to promotes ES cell differentation into the mesoderm lineage.

Project description:We have determined the genome-wide binding profile of the transcription factor Sp5. Flag-tagged Sp5 was targeted to the HPRT locus in A2Lox.cre ES cells to allow for Doxycycline inducible expression. ES cells were cultured as embryoid bodies for 2 days to prime them for germ layer differentiation. Cells were then treated with Doxycycline for 24 hrs. We found that Flag-Sp5 prefentially bound to promoters associatd with several growth factor signalling pathways, but most prominently with the Wnt/beta-catenin pathway to selectively promote mesoderm differentiation over neural. These data implicate Sp5 as new regulator of Wnt/beta-catenin target expression to promotes ES cell differentation into the mesoderm lineage. Project description:Lef1 is a critical transducer of the Wnt/beta-catenin signaling pathway that is essential for mesoderm differentiation in vertebrate embryos. We established a doxycycline inducible ES cell line to over-express Flag-tagged Lef1 in differentiating ES cells which have the capacity to form mesoderm cells in vitro. The goal of this study was to identify the genes/gene networks regulated by Lef1 to understand how it regulates mesoderm differentiation and to establish the Lef1-regulated, Wnt/beta-catenin transcriptome. Total RNA was extracted from ES cells after 3 days of differentiating following Lef1 over-expression from day 2 to 3. We compared the transcriptome of differentially expressed genes in untreated and doxycycline treated ES cells to identify Lef1 regulated genes.

Transcription Factors Sp8 and Sp9 Coordinately Regulate Olfactory Bulb Interneuron Development [ChIP-Seq]

Project description:We conducted RNA-Seq and in situ hybridization to identify transcriptional profile changes in the Sp8/Sp9-DCKO olfactory bulb. Simultaneously, we did ChIP-Seq with homemade Sp9 antibody to identify Sp9 binding motifs and which changed genes might be the direct targets of transcription factor Sp9. We found that Sp9 directly binded to key genes which are required for olfactory bulb development. Transcription factors Sp8 and Sp9 belong to the same transcription factor family, so we think Sp8 and Sp9 have very similar binding sites of transcriptional regulation.

FOXP1 is a transcriptional Coactivator of Wnt/β-catenin signaling in Diffuse Large B-cell Lymphoma

Project description:We used a mass spectrometry-coupled lentiviral ‘CD-tagging’ mutagenesis approach to identify genes that activate Wnt/β-catenin signaling. Human A375 melanoma cells containing a β-catenin-driven GFP (green fluorescent protein) transcriptional reporter were transduced with CDBF lentivirus. When integrated near an expressed and spliced gene, the cytomegalovirus (CMV) promoter of the CDBF vector drives constitutive BFP (blue fluorescence protein) expression and by virtue of the splice donor (SD) sequence, an overexpressed FLAG-tagged fusion of the targeted gene. Depending on where within the gene locus the CDBP vector integrates, the resulting overexpressed gene product may be full length or amino-terminal trunctated. Fluorescence activated cell sorting (FACS) was used to isolate BFP+/GFP+ (Wnt active) or BFP+/GFP- (Wnt inactive) A375 cells. We reasoned that if successful, FLAG epitope tag immunopurification and mass spectrometry-based identification of the overexpressed fusion proteins would be cheaper, faster and provide more information than traditional PCR-based detection. Five FLAG immunopurification followed by a series of high salt washes, on-bead tryptic digestion and shotgun mass spectrometry (MS) identified 20 high-confidence proteins specific to Wnt-active cells. The high-salt washes removed associated proteins from the FLAG-tagged bait proteins. The FOXP1 transcription factor ranked as the top screen hit, as determined by spectral count abundance and the CompPASS WD-score.
