Effect of hypoxia on murine splenic NK cells

ABSTRACT: NK cells responses can be significantly affected by microenvironmental conditions. In pathological conditions, oxygen concentration in tissues can be reduced, lading to hypoxia. Here, we investigated how prolonged exposure to hypoxia affects the transcriptome of murine splenic NK cells.

Project description:NK cells responses can be significantly affected by microenvironmental conditions. In pathological conditions, oxygen concentration in tissues can be reduced, lading to hypoxia. Cells adapt to hypoxia by stabilizing transcription factor HIF-1a that mediates its regulatory functions by modifying gene transcription. In many cells, HIF-1a pathway intersect with the activation of the transcription factor AhR due to shared transcriptional co-activator. Here, we investigated how HIF-1a and AhR regulate murine NK cell transcriptome under hypoxia.

Transcriptome of mouse splenic NK cells with MYC deficiency

Project description:To understand the mechanism of the defective NK cell functions with MYC deficiency, we isolated splenic NK cells (CD3-NK1.1+CD49b+) from Mycf/f and MycΔ/Δ/Ncr1Cre mice and performed RNA-seq analysis. It revealed that ribosome and natural killer cell-mediated cytotoxicity were the two most enriched pathways in the differentially down-regulated genes. Functional studies showed reduced synthesis of NK cell effector molecules in the NK Cells with MYC deficiency. Collectively, these data suggest impaired ribosomal biogenesis and reduced synthesis of cytotoxic molecule in the NK cells with MYC deficiency.

Gene expression profile of isolated splenic NK cells from mice treated with ptaquiloside and/or selenium

Project description:To better understand the immunosuppressor mechanism of ptaquiloside in splenic NK cells and the reversion of this effect by selenium, we have employed whole genome microarray expression profile to identify genes associated with immunosuppression. Among 89 genes induced by ptaquiloside treatment in splenic NK cells only two genes (Mt1 and Mt2) were identified as related with its immunosuppressor effect. Moreover this augmented expression of Mt1 and Mt2 was totally abrogated by selenium co-treatment. These results were confirmed by flow cytometry in splenic cells harvested from other six mice and treated in vitro for 1 hour with ptaquiloside and/or selenium. Twenty mice were separated randomly into four groups as Control (water), Pt (ptaquiloside 5.3 mg/kg), PtSe (ptaquiloside 5.3 mg/kg and selenium 1.3 mg/kg) and Se (selenium 1.3 mg/kg) and were treated daily by gavage for 14 days. After treatment, untouched NK cells were isolated using the NK cell isolation kit, LS columns, and QuadroMACS cell separator system (Miltenyi Biotec, Inc.) to perform RNA isolation and whole-genome gene expression profile. Thereby, five independent experiments were performed per group using different donors for each experiment. To confirm the increase of metallothionein protein induced by ptaquiloside, splenic cells were harvested from other six mice and treated in vitro for 1 hour with ptaquiloside [4.4 M-BM-5g/mL] and/or selenium [0.1 mM] and analyzed by flow cytometry.

Tissue-Resident Natural Killer (NK) Cells Are Cell Lineages Distinct From Thymic and Conventional Splenic NK Cells (part 1)

Project description:Natural killer (NK) cells belong to the innate immune system where they can control virus infections and developing tumors by cytotoxicity and production of inflammatory cytokines. Most studies of mouse NK cells, however, have focused on conventional NK (cNK) cells found in the spleen. Recently, we described two populations of NK cells within the liver, tissue-resident NK (trNK) cells and those resembling splenic cNK cells. However, the lineage relationship of trNK to cNK cells was unclear because trNK cells display a phenotype associated with immature, developing cNK cells. Moreover, liver trNK cells could be related to thymic NK cells or alternatively, a lineage distinct from both cNK and thymic NK cells. Herein we used detailed transcriptomic, flow cytometric, and functional analysis of mice deficient in several transcription factors to determine that liver trNK cells form a distinct lineage from cNK and thymic NK cells, especially because they do not require NFIL3 (E4BP4), the previously described NK cellspecification factor. Analysis of other tissues indicate the presence of trNK cells in skin and uterus with different transcription factor requirements. Thus, there are at least four distinct lineages of NK cells: cNK, thymic, liver (and skin) trNK, and uterine trNK cells. Liver NK 1.1+CD49+, liver NK 1.1+CD49-, spleen NK 1.1+ CD49- populations of NK cells were sorted with FACS pooling cells from individual mice to end up with ~100k cells for each samples. mRNA was derived from lysates using Invitrogen oligo-dT beads
