RNA-seq of PgrCre/+Srfflox/flox and Srfflox/flox mouse uterus at gestation day 3.5

ABSTRACT: Upon conditional ablation of Srf in the Pgr-expressing cells of the mouse uterus, PgrCre/+Srff/f (SrfKO) females were infertile, displaying uterine fibrosis and failure to decidualize or support embryo implantation. RNA-seq analysis demonstrated dramatic changes in gestation day 3.5 SrfKO uterine tissues including dysregulation of cytoskeletal remodeling and growth response genes in addition to strong induction of proinflammatory cytokine signaling. These data show that SRF is key for endometrial function and fertility through regulation of inflammatory homeostasis.

Project description:Single cell RNA-seq analysis demonstrated dramatic changes in adult PgrCre/+Srfflox/flox endometrial cell populations compared to Srf flox/flox controls. Uterine Srf ablation caused myeloid immune cell infiltration, and differential expression analysis demonstrated that PgrCre/+Srfflox/flox stromal fibroblasts downregulated progesterone-regulated genes and growth response genes. Proliferating fibroblast populations shifted from G2-M phase-dominant in controls to S-G2 phase-dominant in PgrCre/+Srfflox/flox uteri. PgrCre/+Srfflox/flox epithelial cells overexpressed estrogenic and innate inflammatory genes.

Project description:Chromatin immunoprecipitation for the progesterone receptor (PGR) and sequencing was performed on Pgrcre/-mPRALsL/+ mice. These mice exhibit no endogenous PGR expression, yet exhibit constitutive expression of the PGR-A isoform in PGR positive tissues due to the presence of a conditional expression allele. Mice were ovariectomized, rested for two weeks, and treated with progesterone for 1 hour before harvesting and freezing whole uterine tissue.

Project description:Estrogen (E2) signaling through its nuclear receptor, estrogen receptor α (ERα) increases insulin-like growth factor 1 (IGF1) in the rodent uterus, which then initiates further signals via the IGF1 receptor (IGF1R). Directly administering IGF1 results in similar biological and transcriptional uterine responses. Our studies using global ERα-null mice demonstrated a loss of uterine biological responses of the uterus to E2 or IGF1 treatment, while maintaining transcriptional responses to IGF1. To address this discrepancy in the need for uterine ERα in mediating the IGF1 transcriptional vs. growth responses, we assessed the IGF1 transcriptional responses in PgrCre+Esr1f/f (called ERαUtcKO) mice, which selectively lack ERα in progesterone receptor (PGR) expressing cells, including all uterine cells, while maintaining ERα expression in other tissues and cells that do not express Pgr. Additionally, we profiled IGF1-induced ERα binding sites in uterine chromatin using ChIP-seq. Herein, we explore the transcriptional and molecular signaling that underlies our findings to refine our understanding of uterine IGF1 signaling and identify ERα-mediated and ERα-independent uterine transcriptional responses. Defining these mechanisms in vivo in whole tissue and animal contexts provides details of nuclear receptor mediated mechanisms that impact biological systems and have potential applicability to reproductive processes of humans, livestock and wildlife.

Genome-wide Profiling of FOXA2 and POL2 binding in the mouse uterus [ChIP-Seq]

Project description:Forkhead box A2 (FOXA2) is a critical regulator of endometrial gland development in mice. In the adult mouse uterus, FOXA2 is expressed solely in the GE cells of the endometrium. Conditional deletion of Foxa2 after birth in the uterus, using the progesterone receptor Cre mouse (PgrCre), impeded gland development, thereby rendering the adult mouse infertile due to defects in blastocyst implantation stemming from a lack of endometrial glands and their secretions. As a first step to begin understanding the FOXA2 function in the endometrial glands of the uterus, genome-wide investigation of in vivo FOXA2 and RNA polymerase II (POL2) binding target regions in the neonatal and adult uterus was determined by chromatin immunoprecipitation followed by massively parallel sequencing (ChIP-Seq). In order to determine the transcriptional regulatory networks mediating FOXA2 regulation of endometrial gland development and function, chromatin immunoprecipitation and massively parallel sequencing (ChIP-Seq) was used to create a genome-wide profile of in vivo FOXA2-binding sites in the developing (PD 12) and adult (DOPP 2.5 and 3.5) mouse uterus.

Loss of GRB2 results in nonreceptive endometrium and female infertility.

Project description:To understand the role of GRB2 in the uterus, we generated mice with conditional ablation of Grb2 in the PGR positive cells (Pgrcre/+Grb2f/f; Grb2d/d). Grb2d/d mice are infertile due to implantation failure. Although ovarian functions are normal, Grb2d/d mice had non-receptive endometrium due to dysregulation of progesterone and estrogen signaling. Our transcriptomic analysis identified deficiency of progesterone and FOXA2 signaling.
