Geniposide suppressed tumor progression through DUOX1-mediated ferroptosis in hepatocellular carcinoma

ABSTRACT: Hepatocellular carcinoma (HCC) poses a formidable challenge within the spectrum of digestive system cancers due to its poor prognosis. Geniposide, an iridoid glucoside extracted from the fruits of Gardenia jasminoides Ellis, exhibits a diverse array of biological activities. The objective of this study is to delineate the specific roles and underlying mechanisms of geniposide on the progression of HCC.

ACKR3 Degradation by the Novel Scutellarein Derivative TBS6b Potently Suppresses Hepatocellular Carcinoma

Project description:Hepatocellular carcinoma (HCC) remains a formidable challenge in oncology due to its aggressive nature and limited treatment options. Scutellarein, derived from the traditional Chinese herb Scutellaria baicalensis, has shown potential against HCC, albeit with limited efficacy. To enhance its therapeutic potential, we synthesized a novel scutellarein derivative, TBS6b, incorporating the antitumor active moieties trimethoxyphenyl and benzimidazole. TBS6b exhibited significantly enhanced anti-HCC activity, surpassing its precursor in both in vitro and in vivo models. Comprehensive assays, including colony formation, EdU incorporation, wound healing, and Transwell migration, demonstrated TBS6b's potent inhibition of HCC proliferation, migration, and invasion. Notably, TBS6b's mechanism of action was elucidated as promoting the ubiquitination and subsequent degradation of the atypical chemokine receptor 3 (ACKR3), a key regulator of HCC's aggressive behavior. Rescue experiments further confirmed that ACKR3 downregulation is central to TBS6b’s anti-cancer efficacy. This study not only identifies ACKR3 as a novel therapeutic target in HCC but also introduces TBS6b as a promising candidate for HCC treatment, offering a new avenue for therapeutic intervention in this lethal disease.

Project description:The goal of this study was to delineate the important EZH2 direct target genes that mediate the oncogenic properties of EZH2 in HCC. The EZH2 direct target genes in two HCC cell lines were identified by chromatin immunoprecipitation microarray (ChIP-chip) analysis and later confirmed by independent ChIP-PCR. The functions of the target genes were further examined. Two human HCC cell lines, Huh7 and PLC/PRF/5 (PLC5), were grown in DMEM supplemented with 10% fetal bovine serum. ChIP assays were performed using anti-EZH2 antibody. The immunoprecipitated and input DNA were used to probe the Agilent human ChIP-chip arrays.

An Esterase-Responsive SLC7A11 shRNA Delivery System Induced Ferroptosis and Suppressed Hepatocellular Carcinoma Progression.

Project description:Ferroptosis has garnered attention as a potential approach to fight against cancer, which is characterized by the iron-driven buildup of lipid peroxidation. However, the robust defense mechanisms against intracellular ferroptosis pose significant challenges to its effective induction. In this paper, an effective gene delivery vehicle was developed to transport solute carrier family 7 member 11 (SLC7A11) shRNA (shSLC7A11), which downregulates the expression of the channel protein SLC7A11 and glutathione peroxidase 4 (GPX4), evoking a surge in reactive oxygen species production, iron accumulation, and lipid peroxidation in hepatocellular carcinoma (HCC) cells, and subsequently leading to ferroptosis. This delivery system is composed of an HCC-targeting lipid layer and esterase-responsive cationic polymer, a poly{N-[2-(acryloyloxy)ethyl]-N-[p-acetyloxyphenyl]-N} (PQDEA) condensed shSLC7A11 core (G-LPQDEA/shSLC7A11). After intravenous (i.v.) injection, G-LPQDEA/shSLC7A11 quickly accumulated in the tumor, retarding its growth by 77% and improving survival by two times. This study is the first to construct a gene delivery system, G-LPQDEA/shSLC7A11, that effectively inhibits HCC progression by downregulating SLC7A11 expression. This underscores its therapeutic potential as a safe and valuable candidate for clinical treatment.

Circular RNA Expression is Suppressed by Androgen Receptor (AR) Regulated Adenosine DeAminase that act on RNA (ADAR1) in Human Hepatocellular Carcinoma

Project description:Hepatocellular carcinoma (HCC) is a heterogeneous malignancy as a result of complex genetic and epigenetic alterations. HCC is characterized by a clear gender disparity for which lacking a clear mechanistic understanding. By utilizing circular RNA (circRNA) microarray survey coupled with in vitro analysis, we analyzed the influence of androgen receptor (AR) on circRNA expression in HCC. Our results indicated that AR could suppress circRNA expression by up-regulating adenosine deaminase that act on RNA (ADAR1) p110 at a transcriptional level. Reporter assay further confirmed the role of AR as a transcriptional activator of ADAR1 expression. Furthermore, data collected from our center strongly suggest that ADAR1 expression can effectively predict HCC patients’ prognosis and an abnormal overexpression of ADAR1 is positively correlated with AR in HCC. In addition, we found CircARSP91 (hsa_circ_0085154), one of the circRNAs down-regulated by AR in an ADAR1-dependent manner, could inhibit HCC tumor growth both in vitro and in vivo. These findings highlight the fact that AR as a contributing factor for gender disparity in HCC can cause complex consequences though regulation of circRNA expression. Better understanding of roles of circRNA during HCC initiation and progression will be a key to develop novel HCC therapies.

USP22 promotes the proliferation and inhibits Sorafenib-induced ferroptosis of hepatocellular carcinoma cells through its deubiquitinase activity

Project description:Hepatocellular carcinoma (HCC) remains one of the most lethal cancers, characterized by poor prognosis and low life expectancy. Unfortunately, there are very few molecular therapeutic options available for advanced HCC. Sorafenib is a current standard first-line treatment for advanced HCC, but drug resistance significantly limits its therapeutic efficacy. In this study, we identified ubiquitin-specific protease 22 (USP22) as the key regulator of HCC development and Sorafenib resistance. Analysis of TCGA databases revealed that USP22 is highly expressed in HCC tissues and is closely associated with poor patient prognosis. Our data further indicated that USP22 promotes the proliferation of HCC cells via deubiquitinating and stabilizing cyclin-dependent kinase 11B (CDK11B). Additionally, USP22 acts as a novel inducer of Sorafenib resistance and suppresses Sorafenib-triggered ferroptosis in HCC cells. It reduces the transcription of the transferrin receptor (TFRC) by decreasing H2BK120ub occupancy at TFRC transcription start site (TSS) downstream region, thereby inhibiting ferroptosis upon Sorafenib treatment. Finally, animal experiments confirmed the role of USP22 in promoting HCC cell growth and Sorafenib resistance in vivo. Taken together, our findings suggest that USP22 represents a promising prognostic biomarker and therapeutic target for HCC patients, particularly those with Sorafenib resistance.

CRISPR screens uncover protective effect of PSTK to targeted therapy induced ferroptosis in hepatocellular carcinoma

Project description:Hepatocellular carcinoma (HCC) is one of the most frequent malignancies with extremely poor prognosis. Clinical efficacies of existing targeted therapy strategies were not satisfied due to targeted therapy resistance. In this study, we performed a series of CRISPR/Cas9 screens to identify synthetic lethality genes to improve the clinical response of HCC. CRISPR loss-of-function genetic screens were performed to identify synthetic lethality genes of targeted therapies (Abemaciclib, Palbociclib, Sorafenib, Lenvatinib, Regorafenib and Erastin) in HCC cell lines. RNA-seq analysis were performed to clarify potential mechanism. We identified that inhibition of phosphoseryl-tRNA kinase (PSTK) sensitized HCC cells to targeted therapies. PSTK contributes to resistance of targeted therapy induced ferroptosis.
