Analysis of nascent transcription by PRO-cap in HUDEP-2 cells

ABSTRACT: CRISPR editing of an intronic enhancer within the BCL11A gene reactivates fetal hemoglobin (HbF) in adult erythroid cells, offering a potential gene-based therapy for hemoglobinopathies. However, the mechanisms underlying the remarkable efficacy of CRISPR-mediated enhancer ablation remain poorly understood. Enhancer RNAs, transcribed from enhancers, play a role in gene regulation. We apply PRO-cap to map the location of active polymerases and transcription start sites.
