Single-Cell Transcriptome Profiling of PBMCs in Response to Diverse Immune Adjuvant Stimuli

ABSTRACT: Immune adjuvants are agents that enhance the immunogenicity of vaccines when co-administered with antigens. While their primary mode of action involves activation of innate immune pathways in antigen-presenting cells such as myeloid cells—leading to cellular maturation and enhanced antigen presentation—how adjuvants affect other hematopoietic cell types remains largely unclear. To address this, we performed single-cell RNA sequencing of human PBMCs treated with various immune adjuvants, aiming to characterize cell type–specific transcriptional responses at single-cell resolution.

Combination adjuvant improves influenza virus immunity by downregulation of immune homeostasis genes in lymphocytes

Project description:Adjuvants play a central role in enhancing the immunogenicity of otherwise poorly immunogenic vaccine antigens. Combining adjuvants has the potential to enhance vaccine immunogenicity compared with single adjuvants, although the cellular and molecular mechanisms of combination adjuvants are not well understood. Using the influenza virus hemagglutinin H5 antigen, we define the immunological landscape of combining CpG and MPLA (TLR-9 and TLR-4 agonists, respectively) with a squalene nanoemulsion (AddaVax) using immunologic and transcriptomic profiling. Mice immunized and boosted with recombinant H5 in AddaVax, CpG+MPLA, or AddaVax plus CpG+MPLA (IVAX-1) produced comparable levels of neutralizing antibodies and were equally well protected against the H5N1 challenge. However, after challenge with H5N1 virus, H5/IVAX-1–immunized mice had 100- to 300-fold lower virus lung titers than mice receiving H5 in AddaVax or CpG+MPLA separately. Consistent with enhanced viral clearance, unsupervised expression analysis of draining lymph node cells revealed the combination adjuvant IVAX-1 significantly downregulated immune homeostasis genes, and induced higher numbers of antibody-producing plasmablasts than either AddaVax or CpG+MPLA. IVAX-1 was also more effective after single-dose administration than either AddaVax or CpG+MPLA. These data reveal a novel molecular framework for understanding the mechanisms of combination adjuvants, such as IVAX-1, and highlight their potential for the development of more effective vaccines against respiratory viruses.

Project description:Vaccine research today is focused on using safer, highly purified or recombinant antigens with poor immunogenicity, which has created a need for potent adjuvants. Rational design of effective and safe mucosal adjuvants for human use necessitates a thorough understanding of the mode of action of successful candidate adjuvants. We used microarray to comprehend the molecular signatures of mucosal adjuvants in the mouse vagina. The adjuvants studied, CpG-ODN and ‚âà√≠¬¨¬±-GalCer have previously been shown to be potent mucosal adjuvants in mice when administrered together with a glycoprotein from HSV-2. Two individual experiments were performed, called ES1 and ES2, each experiment contained 4 groups of mice. All mice were pre-treated with progesteron (DP) before intravaginally recieveing either CpG ODN, alpha-GalCer or their respective buffers, PBS and PBS/Tween. Vaginas were excised at 3 different time-points; 4h, 24h and 48h following adjuvant delivery.

Project description:Nanoparticles and microparticles are widely used as vaccine adjuvants to enhance the immune response and improve the stability of antigens. While aluminum salt is one of the most common adjuvants approved for human use, its immunostimulatory capacity is suboptimal. In this study, we modified risedronate, an immunostimulant and anti‐osteoporotic drug, to create the zinc salt particle-based risedronate (Zn-RS), also termed particulate risedronate. Compared to soluble risedronate, the Zn-RS adjuvant demonstrated increased recruitment of innate cells, enhanced antigen uptake locally, and a similar antigen depot effect as aluminum salt. Furthermore, the Zn-RS adjuvant directly and quickly stimulated immune cells, accelerated the formulation of germinal centers in the lymph nodes, and facilitated the rapid production of antibodies. Importantly, the Zn-RS adjuvant exhibited superior performance in both young and aged mice, effectively protecting against respiratory diseases such as SARS-CoV-2 infection. Consequently, particulate risedronate showed great potential as a universal vaccine adjuvant, particularly beneficial for vaccines targeting the susceptible elderly.

Project description:Antigen uptake, processing and presentation by dendritic cells are regulated by complex intra- and inter-cellular signalling events. Typical vaccine adjuvants lead to the transcription of pro-inflammatory cytokines and chemokines which relate to immune induction. We used microarrays to detail the global expression of genes in BMDC underlying emulsion internalization and DC activation with a non-inflammatory nanoemulsion adjuvant. BMDC were incubated with 0.001% v/v naomeulsion (W805EC) or media alone for 6 hours. Total RNA was extracted per sample using RNA easy (Qiagen).

We used microarrays to detail the global expression of genes in murine nasal mucosa underlying immune induction with a non-inflammatory nanoemulsion nasal adjuvant. 8 week old female CD-1 mice were nasally treated with 5 microliters/nare of either 20% (v/v) naomeulsion (W805EC) or PBS. Nasal epithelium was harvested immediately post-mortem at either 6 (6 hr) or 24 hours (24 hr) following treatment. The tissue was placed in OTC and frozen by immersion in liquid nitrogen. Total RNA was extracted per sample using RNA easy (Qiagen).
