Resveratrol induces downregulation of DNA repair genes in MCF7 human breast cancer cells.

Project description:Breast cancer is a worlwide health problem. One of the factors involved in carcinogenesis is diet. Several studies indicate that resveratrol, a polyphenol present in diet, has several antitumor properties. The purpose of the study is gain an insight of resveratrol effect in cancer by determining the gene expression profile of breast cancer cell lines treated with the compound. MCF7 cell lines were treated for a 48 hour period with two concentrations (150 and 250 uM) of resveratrol. 9 plates were used for each condition. The RNA obtained from 3 plates of the same condition was pooled, processed and hybridized to the Affymetrix Human Gene 1.0 ST array. The arrays were analyzed using Partek Genomics suite software-.

Project description:We used DNA microarrays to investigate the impact of indole and 7-hdroxyindole on P. aeruginosa PAO1.For the microarray experiments, 10 g glass wool (Corning Glass Works, Corning, N.Y.) were used to form biofilms in 250 mL in 1 L Erlenmeyer shake flasks which were inoculated with overnight cultures of P. aeruginosa PAO1 diluted that were 1:100. For P. aeruginosa with 7-hydroxyindole and indole, 500 uM 7-hydroxyindole in 250 uL DMF, 1000 uM indole in 250 uL DMF, or 250 uL DMF alone were added to cells grown in LB. The cells were shaken at 250 rpm and at 37oC for 7 h to form biofilms on the glass wool, and RNA was isolated from the biofilm cells. Project description:To identify molecular effects of the tyrosine kinase inhibitor cabozantinib (XL184), we applied gene expression profiling on human acute leukemic cell lines Kasumi-1, which harbors t(8;21) coupled with KIT mutation, by Whole Transcriptome Shotgun Sequencing (RNA-seq). After Kasumi-1 cells were treated with 100nM cabozantinib for 4h or 24h, differentially expressed genes (DEGs) of 124 genes upregulated and 416 were downregulated in 4h, and 184 genes upregulated and 69 genes were downregulated in 24h treatment, compared with vehicle group.
