Expression data from mouse E15.5 kidneys expressing IlkT173I or IlkWT

ABSTRACT: Mammalian kidney development relies on interaction between three major lineages: the ureteric lineage, the nephrogenic lineage and the stromal lineage. Ilk expression was reported as a regulator of development of the ureteric lineage. We used microarray to analyze gene expression in the ureteric and non-ureteric cells from mouse kidneys expressing IlkT173I, a missense variant of ILK identified from a human patient with Congenital Anomalies of the Kidney and Urinary Tract (CAKUT)

Project description:We analyzed lncRNAs located in CNV loci associated with congenital anomalies of the kidney and urinary tract (CAKUT). HSALNG0134318 at CNV 22q11 was identified as CAKUT related lncRNA, which was coexpressed with multiple known CAKUT associated genes. To validate our findings, we performed knockdown experiments in HEK293 cell line to characterize the trascriptomic profiles regulated by HSALNG0134318 in human embryonic kidney cells.

Project description:Mutations in ZMYM2 lead to syndromic congenital anomalies of the kidney and urinary tract (CAKUT) in humans. Tbx18 is co-expressed with Zmym2 in mesenchymal compartment of developing mouse ureter, indicating a potential in vivo relevance of the TBX18–ZMYM2 protein interaction in ureter development. The presence of multiple phenotypes beyond the urinary system in CAKUT patients carrying ZMYM2 mutations suggests that ZMYM2 has extensive roles in various developmental processes.

Nano-LC-MS/MS proteomics in amniotic fluid from CAKUT patients

Project description:The objective of the present work was to identify new renal proteins responsible for Congenital Anomalies of the Kidney and of the Urinary Tract (CAKUT pathophysiology). We conducted a comparative LC-MS/MS-based proteomics analysis of amniotic fluids (AF) collected from non-severe CAKUT (ns-CAKUT; n=19), severe CAKUT (s-CAKUT; n=14), and healthy controls (cont; n=22) fetuses. We identified a total of 224 gestational age independent proteins that were significantly different in abundance when comparing (one-way ANOVA corrected p<0.05) the 3 groups of amniotic fluids 2 by 2. We identified 8 gestational-age independent proteins that were in common when comparing the 3 groups two by two, and that were considered as associated to both CAKUT onset and CAKUT severity.

The Essential Role of ASH2L in Regulating Cell Cycle-Associated Gene Expression to Promote Nephron Progenitor Self-Renewal and Nephron Formation in Mice [RNA-seq]

Project description:Maintaining adequate nephron number is crucial for normal kidney function, as even slight deficits can predispose individuals to hypertension and other late-onset renal diseases. Congenital anomalies of the kidney and urinary tract arise from abnormal embryonic kidney development, which involves the coordinated reciprocal induction of the ureteric bud and metanephric mesenchyme. Preserving the balance between self-renewal and differentiation of nephron progenitor cells in the metanephric mesenchyme is essential for nephron endowment. Prior work has implicated histone modifications in regulating kidney lineage-specific gene transcription and nephron endowment. Here, we selectively inactivated the H3K4 methyltransferase complex component ASH2L in mouse nephron progenitor cells to elucidate its role in nephron progenitor cell self-renewal and differentiation. The results showed that Ash2l inactivation disrupted H3K4 trimethylation establishment at promoters of genes controlling nephron progenitor cell stemness, differentiation, and cell cycle, leading to premature nephron progenitor cell depletion, cell cycle arrest, and abnormal differentiation of early nephron progenitors, resulting in renal dysplasia at birth. These findings identify ASH2L-mediated H3K4 methylation as a critical epigenetic regulator of kidney development and suggest a novel mechanism contributing to congenital anomalies of the kidney and urinary tract. This knowledge may guide future efforts to unravel the pathogenesis of these conditions and develop targeted therapies.

Single cell transcriptomic analysis of nephron and UB lineages in recombinant hPSC-derived kidney organoids grown under various conditions.

Project description:To model tissue interactions in kidney development, a protocol was devised to combine human pluripotent stem cell (hPSC)-derived ureteric bud progenitors with nephrogenic mesenchyme to form kidney organoids. The resulting organoids had a complex composition including the nephron, ureteric, stromal, and endothelial lineages. scRNA-seq was performed to catalog and analyze the populations and developmental trajectories of the different lineages.
