Gene expression profile of tumour-infiltrating immune cells in murine PDAC

ABSTRACT: We established an organoid-based murine pancreatic cancer model that closely resembles human PDAC. To characterize the tumor-infiltrating immune cell states following combination therapy with immune checkpoint inhibitors (ICIs) targeting PD-1 and CTLA-4, along with agonistic CD40 antibodies (aCD40), we performed RNA sequencing (RNA-seq) on tumor-infiltrating CD45+ cells from our model.

Bispecific antibodies increase the therapeutic window of CD40 agonists through selective dendritic cell targeting

Project description:Therapeutic use of agonistic anti-CD40 antibodies is a potentially powerful approach for activating the immune response to eradicate tumors. However, the translation of this approach to clinical practice has been significantly restricted due to the severe dose-limiting toxicities observed in multiple clinical trials. Here, we demonstrate that conventional type-1 dendritic cells are essential for triggering antitumor immunity but not toxicity by CD40 agonists, while macrophages, platelets, and monocytes lead to the toxic events. Therefore, we designed bispecific antibodies that target CD40 activation preferentially to dendritic cells. These bispecific reagents demonstrate a superior safety profile compared to their parental CD40 monospecific antibody, while triggering potent anti-tumor activity. We suggest such cell-selective bispecific agonistic antibodies as a drug platform to bypass the dose-limiting toxicities of anti-CD40, and of additional types of agonistic antibodies used for cancer immunotherapy.

Inflammatory Conditions Promote Resistance to Immune Checkpoint Inhibitors in High Microsatellite Instability Colorectal Cancer

Project description:Inflammatory conditions caused by obstruction or perforation are common complications in colorectal cancer (CRC) and play important roles in tumor progression and immunosuppression. However, the significance of inflammatory conditions in the tumor response to immune checkpoint inhibitors (ICIs) remains unclear. We found a high microsatellite instability (MSI-H) CRC patient (Patient 1) whose primary tumor progressed and liver metastasis regressed during PD-1 blockade after having inflammatory conditions. Then, in 73 MSI-H CRCs, inflammatory conditions during ICI treatment were correlated with a poor tumor response, and an elevated NLR was associated with a poor immune status and resistance to ICIs. An organoid-T cell coculture model demonstrated an inhibited local immune response to treatment instead of systemic immunosuppression in Patient 1. Single-cell RNA sequencing suggested that neutrophils suppress the immune microenvironment mostly through CTLA-4-associated pathways. Therefore, inflammatory conditions in MSI-H CRCs correlate with resistance to ICIs through neutrophil-associated immunosuppression. Additional CTLA-4 blockade may improve the sensitivity to PD-1 blockade.

T cells and monocyte-derived myeloid cells mediate immunotherapy-related hepatitis in a mouse model

Project description:Immune checkpoint inhibitors (ICIs) are associated with immune-related adverse events (irAEs) which are more severe when ICIs are used in combination. A mouse model was developed to elucidate the molecular mechanisms of immune-related hepatitis, one of the common irAEs associated with ICIs. Molecular profiling by single cell RNA sequencing was performed on Pdcd1-/- mice treated with anti-CTLA4 and/or the IDO1 inhibitor epacadostat or a 4-1BB agonistic antibody. ICI combination-induced hepatitis and 4-1BB agonist-mediated hepatitis share similar features yet maintain distinct immune signatures. Both were characterized by an expansion of activated T cells. Single-cell transcriptomics revealed that the hepatitis induced by combination ICIs is associated with a robust immune activation signature in all subtypes of T cells and T helper 1 skewing. Expression profiling revealed a central role for IFNγ and liver monocyte-derived macrophages in promoting a pro-inflammatory T-cell response to ICI combination and 4-1BB agonism.
