Malignant ascites is associated with PIGR+RORC+ dendritic cell subset

ABSTRACT: Malignant ascites is caused by the peritoneal dissemination of cancer and associated with a poor prognosis. We sought to identify unique features of the ascites microenvironment and leveraged single-cell RNA sequencing in patients with ascites from gastroesophageal adenocarcinoma. We identified a dendritic cell (DC) subset that expresses RORC and foundt that this subset exists in low abundance in human tissues but is enriched in malignant ascites from multiple tumor histologies. We isolated and characterized this cell subset as a DC, and the expression of genes that define this subset can be induced by ascites supernatant. This DC subset appears to be associated with the peritoneal spread of cancer across tumor types and may help to provide opportunities to understand therapeutic resistance in the peritoneal cavity.
