Transcriptome profiling of human PDLSC under GM/OM with BMP2/BMP9 at days 1, 7, and 15

ABSTRACT: We investigated the gene expression changes in human periodontal ligament stem cells (PDLSCs) cultured in growth medium (GM) or osteogenic medium (OM) with various treatments (NT, BMP2, BMP9) over three time points (1d, 7d, 15d). Each group has two biological replicates. Total 12 groups, 24 samples. Affymetrix Human Gene 2.0 ST arrays were used.

BMP9 Secretome of hHiPC LC-MS/MS

Project description:To investigate BMP9/ALK1 signaling in human highly proliferative cells (hHiPC), we selected one primary population of hHiPC from 3 individuals and studied the secretome responses to BMP9 treatment. Proteins were collected in hHiPC treated conditioned media (CM) for mass spectrometry analysis. Proteomic analysis of the hHiPC secretome following BMP9 treatment demonstrates that the secreted proteins, sclerostin (SOST), meflin/immunoglobulin superfamily containing leucine rich repeat (ISLR), and insulin-like growth factor binding protein-3 (IGFBP3), are novel downstream targets of BMP9/ALK1-mediated signaling. Consistent with this hypothesis, lentiviral shRNA and pharmacological inhibition of ALK1 in hHiPCs suppressed secretion of SOST, ISLR, and IGFBP3 following BMP9 treatment as measured by secretome LC-MS/MS.

Subconfluent mouse C3H10T1/2 cells were transduced with adenoviral vector expressing BMP9 or GFP for 30h. Total RNA was isolated for microarray analysis using Affymetrix mouse MOE430A GeneChips. Experiments were done in triplicate.

BMP9 signaling promotes the normalization of tumor blood vessels

Project description:We sought to evaluate the effects of BMP9 on tumor vascular formation. We observed that BMP9 promotes vascular normalization with decreased permeability and increased perfusion and perivascular cell coverage. Gene expression analysis of CD31-positive cells isolated from control and BMP9-overexpressing tumors revealed enrichment of pathways involved in Notch signaling and vascular quiescence.

IdentiM-oM-,M-^Acation of genes responsible for biosynthesis of kojic acid in Aspergillus oryzae

Project description:Kojic acid is produced in large amounts by Aspergillus oryzae as a secondary metabolite and is widely used in the cosmetic industry. Glucose can be converted to kojic acid, perhaps by only a few steps, but no genes for the conversion have thus far been revealed. Using a DNA microarray, gene expression proM-oM-,M-^Ales under three pairs of conditions signiM-oM-,M-^Acantly affecting kojic acid production were compared. All genes were ranked using an index parameter reM-oM-,M-^Becting both high amounts of transcription and a high induction ratio under producing conditions. After disruption of nine candidate genes selected from the top of the list, two genes of unknown function were found to be responsible for kojic acid biosynthesis, one having an oxidoreductase motif and the other a transporter motif. These two genes are closely associated in the genome, showing typical characteristics of genes involved in secondary metabolism. Analysis in two-color microarray experiments used total RNA isolated from A. oryzae mycelia in three pairs of kojic acid producing to non(less)-producing conditions. The pairs of culture conditions were: 4 d to 2d; 7d to 4d; nitrate(-) to nitrate(+) in kojic acid producing medium.

BMP9 and BMP10 coordinate liver cellular crosstalk to maintain liver health

Project description:The liver is a largest solid organ in the body and is majorly composed of HCs, ECs, KCs, and HSCs, which spatially interact and cooperate each other to maintain liver homeostasis. However, the complexity and molecular mechanisms underlying the crosstalk between these different cell types remain to be revealed. Here, we generated mice with conditional deletion of Bmp9/10 in different liver cell types and demonstrated that HSCs was the major source of BMP9 and BMP10 in the liver. Using transgenic ALK1 (receptor for BMP9/10) reporter mice, we found that ALK1 is expressed on KCs and ECs other than HCs and HSCs. KCs from Bmp9/10HSC-KO (conditional deletion of Bmp9/10 from HSCs) mice lost their signature genes expression, such as ID1/3, CLEC4F, VSIG4 and CLEC2, and were replaced by monocyte-derived macrophages. ECs from Bmp9/10HSC-KO mice also lost their identity and were transdifferentiated to continuous ECs, ultimately leading to collagen IV deposition and liver fibrosis. Hepatic ECs expressed several angiocrine factors, such as BMP2, BMP6, Wnt2 and Rspo3, to regulate liver iron metabolism and metabolic zonation. We found that these angiocrine factors were significantly decreased in ECs from Bmp9/10HSC-KO mice, which further resulted in liver iron overload and disruption of HCs zonation. In addition, focal fatty liver spontaneously occurred in Bmp9/10HSC-KO mice at the age of 28W. In summary, we demonstrated that HSCs play a central role in mediating liver cell-cell crosstalk via production of BMP9/10 to maintain liver health.
