The different genes expression of PLC8024 cell after drug treatment

ABSTRACT: Liver cancer is a highly malignant tumor. Current treatments are not effective. Hence, more effective treatment methods need to be sought. We aim to identify different genes after drug treatment by RNA-seq, and subsequently explore their functions, in the hope of finding better targets for liver cancer treatment.

Project description:The pressure changes in hepatic microenvironment may be involved in the regulation of proliferation and invasion of hepatocellular carcinoma, but the mechanism is unknown. This project simulates the mechanical microenvironmental conditions of liver cirrhosis, exerts the static water pressure on human hepatoma cell lines and observes the changes of proliferation and invasion of hepatoma cells under different pressure loading conditions. Then screens the differential expression mechanic-responsive miRNAs under the optimal loading condition and carry out biological verification. Bioinformatics analysis was used to explore the related pathways. This study provides a theoretical basis for the prevention and treatment of liver cancer and provides a direction for finding effective drugs for clinical treatment of liver cancer.

Hypothermic oxygenated machine perfusion influences the immunogenicity of donor livers in humans

Project description:Hypothermic oxygenated machine perfusion (HOPE) is an organ preservation strategy shown to reduce ischemia-reperfusion-injury (IRI)-related complications following liver transplantation this remains to be evaluated in humans. Analysis of liver tissue transcriptome at reperfusion revealed an effect of HOPE on the reactive oxygen species pathway. Two weeks post transplantation, HOPE recipients exhibited increased circulating CD4+FOXP3+CD127lo regulatory T cells (Tregs) (p<0.01), which corresponded to a higher frequency of donor specific Tregs (p<0.01) and was followed by reduced alloreactivity index of CD8+ T cells 3 months post-transplant. Our study provides novel mechanistic insight into the capacity of HOPE to influence liver IRI and to modulate effector and regulatory donor-specific Tcell responses post-transplantation. These findings, which confirm observations made in animal models, help explain the decreased rejection rates reported in patients receiving HOPE-treated allografts.

Majusculamide-o, a Simplified Marine Natural Product Analog, Exhibits Potent and Specific Cancer Cell Cytotoxicity

Project description:Metastatic solid tumors (e.g., ovary, pancreas, liver, lung, and brain) contribute to a high mortality rate in cancer patients with few therapeutically effective anticancer drugs available for their treatment, highlighting the need to develop agents that target solid tumors. Natural products (NPs) derived from cyanobacteria possess potent anticancer activity, yet most are hard to synthesize and modify to improve therapeutic efficacy. Here, we have efficiently synthesized a simplified analog of the marine NP majusculamide D, majusculamide o (maj-o, 1), that has remarkable potency and selective cytotoxicity towards various metastatic cancer cells. We found that maj-o (1) treatment causes apoptosis in various cancer cell lines of the ovary (OVCAR3), pancreas (PANC1), brain (U251N), and lung (H125) in a dose dependent manner with the least cytotoxic effect towards non-metastatic or primary tumors of the liver (HEPG2) and ovary (OVCAR8). 1 significantly alters gene expression signatures with more treatment time and affect pathway associated with PI3K-Akt signaling. Our finding suggests that maj-o has great potential to serve a lead compound for drug discovery of novel antineoplastics for solid tumors.

Drug-tolerant cell-derived exosomes regulate proteasome inhibitor resistance pathways in MLL leukemias

Project description:The interplay between cancer cells and microenvironment can influence treatment response and plays a key role in the emergence of drug resistance. Rapidly acquired resistance prevents proteasome inhibitors (PIs) therapies from achieving stable and complete responses. Investigating the underlying mechanisms and developing effective strategies against PI resistance are highly desired in the clinic. Here we uncovered that PI resistance was reversible in MLL leukemia, which consistent with the finding that patients could regain sensitivity to PIs after a drug holiday. Exosomes derived from drug-tolerant cells could transmit PI resistance to sensitive cells via facilitating cell cycle arrest and stemness pathway in MLL leukemia cells. Furthermore, TieDIE algorithm integration analysis of transcriptome and proteome datasets identified candidate exosomal proteins, providing potential therapeutic targets for treating refractory MLL leukemia. Therefore, exosomal regulatory proteins may serve as a predictor and a potential therapeutic target for PI resistance, and inhibiting the secretion of exosomes is a promising strategy for restoring PI resistance in vivo, which provides a paradigm and may also be applied for the treatment of other cancers resulting from chemotherapy relapse.

PPARδ inhibition blocks tumor-induced Bregs and enhances CD40 agonist immunotherapy

Project description:Most cancers are cold tumors characterized by a lack of effector T cells and hence unresponsive to immune checkpoint blockades, highlighting the importance of repairing the failed T cell priming in cancer immunotherapy. CD40 agonists are efficient in eliciting T cell responses and effective in converting cold tumors into hot, their clinical application, however, is limited by the very narrow window between the effective and toxic dosages. Here, we sought to reduce the effective dosage of CD40 agonists by combination therapy. We identified a CD19+CD24hiCD38lowIgDlow/- regulatory B cell (Breg) subset that is increased in tumor-bearing mice and cancer patients, and discovered that PPARδ is highly expressed in these tumor-associated Bregs and required for their proliferation and function. Inhibition of PPARδ acted synergistically with agonist anti-CD40 to enhance antitumor immunity without increasing toxicity in murine melanoma and bladder carcinoma models. Combination therapy with PPARδ inhibitor could reduce anti-CD40 dosage by over 60% to a well-tolerated level with improved or at least not compromised antitumor effects, providing a potential strategy for safely using CD40 agonists in cancer treatment. PPARδ also offers a new target for selectively blocking Breg activity while sparing immunostimulatory B cells that are known to aid antitumor immunotherapy.
