Effect of decitabine on therapeutic response in doxorubicin- and paclitaxel-resistant breast cancer cell lines


ABSTRACT: Breast cancer is the most commonly diagnosed malignancy in women worldwide, yet treatment success is frequently compromised by the development of therapeutic resistance. The complex nature of this resistance necessitates in-depth investigation to identify the mechanisms underlying the resistant phenotype of tumor cells and to develop effective therapeutic strategies. This study details the derivation and comprehensive functional and molecular characterization of doxorubicin- and paclitaxel-resistant breast cancer cell lines representing three distinct subtypes. Additionally, a triple-negative breast cancer (TNBC) cell line was used to explore the potential of the DNA methyltransferase inhibitor decitabine in reversing aberrant DNA methylation patterns associated with chemotherapy resistance. The study also evaluates decitabine's ability to enhance therapeutic efficacy in treatment-resistant TNBC.
