Ionizing radiation induced changes of miRNA expression in A549 lung cancer cells

Project description:We have employed microarray expression profiling as a discovery platform to identify microRNAs induced by radiation. Human A549 lung cancer cells were irradiated (2Gy/day for consistent 3days) and miRNA signature was identified that distinguished between control and radiation treated samples.Irradiated and un-irradiated A549 cells were harvested after 48 hr of treatment and microarray analysis was performed. Expression of five miRNAs (miR-30a, miR-30b, miR-30c, miR-30d, and miR-30d) from this signature was quantified in the same RNA samples by real-time PCR, confirming variability between control and radiation treated A549 cells.

Expression data of mRNA from wild-type A549 cells and microRNA-203 knockout A549 cells during H5N1 infections

Project description:MicroRNA-203 was up-regulated markedly upon H5N1 virus infection. To identify the potential target genes of miR-203, we constructed a miR-203 knockout A549 cell line. Then wild-type and miR-203 knockout A549 cells were mock-infected or infected with H5N1 virus for 48h. The Agilent Whole Human Genome Oligo Microarray was performed to analyze the mRNA expression profiling. Meanwhile, the online tool TargetScanHuman (http://www.targetscan.org/vert_71/) was used to predict biological targets of miR-203. We combined the predicted genes with the genes differentially expressed in wild-type and miR-203 knockout A549 cells, and preliminarily identified some candidate mRNAs. Then more experiments were performed to further verify these target genes, such as dual-luciferase reporter assay, quantitative real-time PCR or Western blot analysis.

Gene expression signatures for MMP19 full length cDNA transfection in A549 cells.

Project description:To further understand the role of MMP19 in IPF, particularly in genotype and phenotype changes, we have employed whole genome microarray expression profiling as a discovery platform to identify the target genes of MMP19 in epithelial cells. Gene expression analysis showed that 8358 genes were differentially expressed at p-value 0.05 level and 748 genes were significantly changed with MMP19 expression, which covers matrix protien and cell proliferation, motality and phenotype changes (EMT). Among them, PTGS2 was revealed as one of the most significantly up-regulated genes. And PTGS2 up-regulation in IPF lungs was verified by immunohistochemical (IHC), qRT-PCR and Western-blot as well. A549 is a typical epithelial cell line from ATCC which are widely used in the lung cell biology studies in vitro. To define the role of MMP19 in epithelial cells, we employed Agilent whole genome gene expression microarray profiling to identify genotype alteration with overexpression of MMP19 to find the target genes of MMP19, further to explore the function of MMP19 in the unique microenironment of IPF
