Single cell transcriptomic analysis of total lung cells from WT and GFI1 KO mice.


ABSTRACT: GFI1 is a transcription factor known to be essential for the hematopoiesis and for the formation of immune cells. Immune cells represent an important part of the lung tissue so we decided to determine the impact of the absence of GFI1 on lung populations, especially immune cells. Lungs from WT or GFI1 KO adult mice were enzymatically dissociated, before being washed and live lung cells were sorted using the ARIA III sorter. The sorted cells were used for gene expression (10x genomics). The sc RNA-seq highlighted differential transcriptomic changes between the 2 conditions, lung cells from GFI1 KO mice and lung cells from WT mice.This experiment allows us to identify specific populations which were absent or over represented in GFI1 KO lungs.
