Exosomal Non-Coding RNAs Drives Multiple Myeloma Progression

ABSTRACT: This study aimed to investigate gender-specific dysregulation of exosomal non-coding RNAs (ncRNAs) and their regulated mRNAs in multiple myeloma (MM). Exosomes were isolated from six MM cell lines (LP1, NCI-H929, JJN-3, U-266, KMS-11, and ANBL-6), bone marrow aspirates from 24 MM patients (8 female and 16 male), and 7 healthy controls. Exosome characterization using NanoSight NS300 confirmed consistent size distribution and concentration across samples. RNA sequencing was performed on exosomal RNA cargo to profile long non-coding RNAs (lncRNAs), microRNAs (miRNAs), and mRNAs, enabling analysis of their regulatory roles in MM pathogenesis.
