Regulation of STK38 by autophagy governs YAP1 activity during paligenosis


ABSTRACT: Paligenosis is a conserved process that enables mature cells to reenter the cell cycle in response to tissue injury, progressing through stages of autodegradation, gene expression changes, and proliferation. The Hippo signaling pathway, particularly YAP1, plays a key role in regulating this process, with Serine/Threonine Kinase 38 (STK38) identified as a key regulator that inhibits YAP1 in uninjured cells but is degraded via autophagy during injury. This degradation of STK38 leads to YAP1 activation, which drives cellular plasticity and proliferation. Additionally, the secretory maturation factor MIST1 helps preserve STK38 function, suggesting a broader, evolutionarily conserved role for autophagy in tissue regeneration.
