Single-cell RNA-seq reveals disparate macrophage responses to intracellular Salmonella

Project description:Intracellular bacterial pathogens can exhibit large heterogeneity in growth rate inside host cells with major consequences for the infection outcome. If and how the host responds to this heterogeneity remains poorly understood. Here, we combined a fluorescent reporter of bacterial cell division with single-cell RNA-seq analysis to study the macrophage response to different intracellular states of the model pathogen Salmonella enterica serovar Typhimurium. The transcriptomes of individual infected macrophages revealed a spectrum of functional host response states to dividing and non-dividing bacteria. Intriguingly, macrophages harboring non-dividing Salmonella display hallmarks of the pro-inflammatory M1 polarization state and differ little from bystander cells, suggesting that non-dividing bacteria evade recognition by intracellular immune receptors. By contrast, macrophages containing dividing bacteria have turned into an anti-inflammatory, M2-like state, as if fast-growing intracellular Salmonella overcome host defense by reprogramming macrophage polarization. Additionally, our clustering approach reveals intermediate host functional states between these extremes. Altogether our data suggest that gene expression variability in infected host cells shapes different cellular environments, some of which may favor a growth arrest of Salmonella facilitating immune evasion and the establishment of a long-term niche; while others allow Salmonella to escape intracellular antimicrobial activity and proliferate.

Salmonella mediated intracellular niche control by SCV size regulation

Project description:Salmonella is a human and animal pathogen causing gastro-enteric diseases worldwide. The key feature of Salmonella infection is its entry into intestinal epithelial cells within a Salmonella-Containing Vacuole (SCV). This original compartment is distinct from empty macropinosomes formed around the infection site. A few minutes after its formation, the SCV increases in size through fusions with the surrounding macropinosomes. On the opposite, Salmonella induces the formation of elongated tubules leading to SCV membrane and volume loss. Later, the SCV can mature into a vacuolar pathogen niche, or be ruptured releasing Salmonella in the host cytosol where the bacteria hyper-replicates. Here, we describe how size control of the early SCV is the main contributor to its stability and consequently determines the Salmonella intracellular niche and growth. We identify the SNAREs required for increasing the SCV size through fusions. We show that this fusion promotes the maintenance of the SCV integrity and the establishment of a vacuolar niche

Salmonella activation of STAT3 signaling by SarA effector promotes intracellular replication and production of IL-10

Project description:Salmonella enterica is an important foodborne pathogen that utilizes secreted effector proteins to manipulate host pathways to facilitate survival and dissemination. Different S. enterica serovars cause disease syndromes ranging from self-limited gastroenteritis to typhoid fever and vary in their repertoire of effectors. We leveraged this natural diversity to identify stm2585, here designated sarA (Salmonella anti-inflammatory response activator), as a Salmonella effector secreted primary by the SPI-2 type III secretion system. SarA is necessary and sufficient to induce STAT3 phosphorylation and IL-10 production, contributing to intracellular replication in vitro and bacterial load at systemic sites in mice. These results demonstrate that Salmonella has evolved effector mechanisms for regulating a host anti-inflammatory signaling pathway important in infection, autoimmunity, and cancer.

Project description:We report the changes in transcriptional landscape that occur under anaerobic conditions in the absence of a DMSO reductase in Salmonella enterica subsp. enterica serovar Typhimurium strain 14028s. By comparing the transcriptome of the parental, wild-type Salmonella to the transcriptome of a ∆dmsABC strain, we identified that there was a significant upregulation of SPI-2 genes in the ∆dmsABC strain.
