ALKBH5 regulates paclitaxel resistance in NSCLC via inhibiting CEMIP-mediated EMT

Project description:N6-methyladenosine (m6A) is the most printed and prevalent mRNA modification, which was verified to be closely correlated with cancer occurrence and progression. As m6A demethlyase, the dysregulation of ALKBH5 was observed in various cancer. However, the role and underlying machinery of ALKBH5 in NSCLC pathogenesis, especially the chemo-resistance was poorly elucidated. The current study indicated that ALKBH5 was decreased during paclitaxel (PTX) resistant process and its down-regulation usually implied poor prognosis of NSCLC patients. Over-expression of ALKBH5 in PTX-resistant cells could suppressed cell proliferation and enhanced chemo-sensitivity. Whereas, knockdown of ALKBH5 exerted opposite effect, which further supported the tumor suppressive role of ALKBH5. Over-expression of ALKBH5 also could reverse the EMT process in PTX-resistant cancer cells. Mechanistically, data of RNA-seq, Real-time PCR and western blotting indicted that CEMIP, also known as KIAA1199, may be the downstream target of ALKBH5. And ALKBH5 could negatively regulated the CEMIP level by decreasing its mRNA stability. Collectively, current data demonstrated that ALKBH5/CEMIP axis modulates the EMT process in NSCLC, which in turn regulates chemo-sensitivity of cancer cells to PTX.

Single-cell RNA-seq profiling of treatment-naïve chemo-refractory and chemo-sensitive ovarian cancer

Project description:This dataset contains single-cell RNA-seq data from treatment-naïve biopsies of nine patients diagnosed with ovarian high-grade serous carcinoma (HGSC), including four chemo-refractory and five chemo-sensitive cancers. Four of the samples (EOC204, EOC115, EOC649, EOC1127) are newly generated, while five samples have been previously published (GSE165897). The data was generated as part of the DECIDER observational trial (NCT04846933) to investigate the molecular drivers of chemo-refractory HGSC. Key findings from the study indicate that chemo-refractory HGSC is associated with reduced interferon type I (IFN-I) activity and enhanced hypoxia pathway activity, while baseline IFN-I pathway activity in chemo-naïve cancer serves as an independent prognostic factor for chemotherapy response.

Chemo- versus immuno-precipitation of G-quadruplex-DNA (G4- DNA): a direct comparison of the efficiency of the antibody BG4 versus the small-molecule ligands TASQs for G4 affinity capture

Project description:The reliability of the BG4-IP-seq protocol, along with the straightforward access to rice genomic DNA, makes this system ideal for comparing the pull-down efficiency of the antibody BG4 versus the small-molecular baits BioTASQ and BioCyTASQ. These two TASQs (for template-assembled synthetic G-quartets) are biotinylated biomimetic ligands that were successfully employed for fishing G4-RNAs out from human cell lysates, in a protocol referred to as G4RP-seq, which belong to the toolbox of seq-based techniques used to interrogate G4-RNA prevalence, comprising rG4-seq, DMS-seq, Keth-seq and SHALiPE-seq. We reasoned that the ability of TASQs to isolate G4-DNA deserves both to be investigated and to be compared to that of BG4. We thus report here on these investigations. which provides the very first comparison of the efficiency of the immuno- versus chemo- precipitation of G4s, under strictly identical experimental conditions.

A single-cell atlas of immune heterogeneity in anti-PD1-treated non-small cell lung cancer

Project description:Anti-PD(L)1 with chemotherapy is a standard of care for non-small cell lung cancer (NSCLC), but a varying degree of response to the same regimen is observed in patients. The tumor immune microenvironment (TIME) plays a key role in response to immunotherapy, and the TIME heterogeneity in association with therapeutic outcome is incompletely understood. Here we prospectively applied single-cell RNA and TCR sequencing to characterize post-neoadjuvant chemo-immunotherapy treatment tumor samples of 234 NSCLC patients. Our study provides a fine-grained dissection of the TIME heterogeneity underlying response to chemo-immunotherapy in NSCLC, thus representing a valuable resource for improved management of NSCLC.
