Project description:Schisandra chinensis fruit extract (SCE) and its active ingredient Schisandrin B (SchB), which are effective in the treatment of vascular diseases, have been known to suppress transforming growth factor β1 (TGF-β1)-mediated Smad activation and myosin light chain (MLC) phosphorylation in vascular smooth muscle cells (VSMCs). However, it is still largely unknown about the pharmacologic effects and mechanisms of SCE and SchB on TGF-β1-induced intracellular signaling pathways in vascular smooth muscle cells (VSMCs).

Transcriptomics of C. chinensis and E. ulmoides treated aged C. elegans

Project description:The nematode Caenorhabditis elegans was treated with extracts of the Traditional Chinese Medicine plants Cuscuta chinensis and Eucommia ulmoides from the L4 stage. In aged worms (on the 7th and 12th day of adulthood) different health parameters were determined. Besides the prolongation of lifespan, we found that C. chinensis improved the swimming behavior, pharyngeal pumping rate, stress resistance, mechanosensation and memory of aged C. elegans. Furthermore, the extract treatment reduced the autofluorescence, which is a known biomarker of ageing. Thus, we concluded that C. chinensis is an overall healthspan enhancer. In contrast, the E. ulmoides extract specifically enhanced the lifespan and stress resistance of aged C. elegans, but did not improve any other health aspect. To reveal the mechanism behind the healthspan enhancing effects of C. chinensis, the transcriptome of treated and untreated C. elegans on the 12th day of adulthood was analysed. For comparison purposes, E. ulmoides treated nematodes were included in the analysis.

The predatory stink bug Arma chinensis (Hemiptera: Pentatomidae) produces a complex proteinaceous venom to overcome caterpillar prey

Project description:Predatory bugs capture prey by injecting venom from their salivary glands using specialized stylets. Understanding venom function has been impeded by a scarcity of knowledge of their venom composition. We therefore examined the proteinaceous components of the salivary venom of the predatory stink bug Arma chinensis (Hemiptera: Pentatomidae). Using gland extracts and venoms from 5th-instar nymphs or adult females, we performed shotgun proteomics combined with venom gland transcriptomics. We found that the venom of A. chinensis comprised a complex suite of over a hundred individual proteins, including oxidoreductases, transferases, hydrolases, ligases, protease inhibitors, and recognition, transport and binding proteins. Besides the uncharacterized proteins, hydrolases such as venom serine proteases, cathepsins, phospholipase A2, phosphatases, nucleases, alpha-amylases, and chitinases constitute the most abundant protein families. However, salivary proteins shared by and unique to other predatory heteropterans were not detected in A. chinensis venom. Injection of the proteinaceous (> 3 kDa) venom fraction of A. chinensis gland extracts or venom into its prey, the larvae of the Oriental armyworm Mythimna separata (Walker, 1865), revealed insecticidal activity against lepidopterans. Our data expands the knowledge of heteropteran salivary proteins and suggests predatory asopine bugs as a novel source for bioinsecticides.

RNA-seq of Taxus chinensis cells

Project description:We reported the application of high-throughput sequencing technology (RNA-seq) for the transcriptome of T. chinensis cells and the transcriptional alternatives of that responded to MeJA were comprehensively and quantitatively assessed with high-throughput sequencing technology (RNA-seq). By sequencing > 29 million reads (200 bp in length) of cDNA from each of MeJA-treated T. chinensis cells at 16 h (T16) and the control (T0), we identified 46,581 transcripts and uncovered 13,469 genes differentially expressed in response to MeJA. We provided functional clues for understanding the regulation mechanisms of MeJA-mediated defense responses and taxol biosynthesis.

Water Deficit Response Differs Between Invasive T. chinensis and T. ramosissima Established in Southern and Northern United States

Project description:Transcriptomic profiling of T. chinensis and T. ramosissima shows responses due to water deficit that are common between the two species and differences that shows their invasiveness originating from southern and northern united states. Several drought related genes that were up-regulated common in both species and transcription factors unique to T.chinensis and T. ramosissima were also found. Gene Ontology classification shows similar functional categories in both the species. Differences in two species due to water deficit were also illustrated in networks constructed from genes enriched in biological processes and molecular functions.

Sex-specific small RNA profiling and target gene expression in dioecious and monoecious Pistacia chinensis

Project description:Pistacia chinensis Bunge is known as dioecious, but we have found wild monoecious individuals. In order to screen the candidate genes which may influence the sex expression or floral phenotypic differences of P. chinensis, the inflorescence buds for different sex types associated with the sex differentiation were selected and tested for small RNA sequencing. Sex-specific differentially expressed small RNA were discovered, combined with real-time PCR data, the regulation patterns of various sex types were first revealed. Our study represents the first detailed analysis of small RNA sequencing, providing more clues for understanding the mechanism of sex determination on P. chinensis.

Gall aphid saliva LC-MS/MS - Proteins Identified from Saliva and Salivary Glands of the Chinese Gall Aphid Schlechtendalia chinensis

Project description:Aphid saliva plays an essential role in the interaction between aphids and their host plants. Several aphid salivary proteins have been identified and analyzed. However, none of the characterized salivary proteins are from galling aphids. Here we analyzed the salivary proteins from the Chinese gall aphid, Schlechtendalia chinensis using LS-MS/MS analysis. A total of 31 proteins were identified directly from secreted saliva collected via artificial diet, and 141 proteins were identified from protein extracts derived from dissected salivary glands. Among these identified proteins, 17 were found in both secreted saliva and dissected salivary glands. In comparison with salivary proteins identified from three other free living aphids, the most striking feature of the salivary protein from S. chinensis is the existence of high proportion of proteins with binding activity, including DNA binding, protein binding, ATP binding and ion binding proteins et al. We speculate that these binding proteins may be involved in induction of gall formation. Our results provide a framework for future research to elucidate the molecular basis for gall induction by S. chinensis.
