RNA profiling of wild-type Osteosarcoma (U2OS) cells and its KO derivative (PERK KO) upon Palbociclib treatment for 4 days

ABSTRACT: Dysregulation of cyclin-dependent kinase 4/6 (CDK4/6) occurs in various cancers. Three CDK4/6 inhibitors are approved for clinical use including Palbociclib (PD). CDK4/6 inhibitors trigger early G1 cell cycle arrest in cells, leading to cellular quiescence while long term inhibition enforces transition to senescence. PERK is a shared Kinase between the Unfolded Protein Response (UPR) and the Integrated Stress Response (ISR). We report that PERK inhibition accelerate Palbociclib induce senescence.
