Evaluation of gene expressions in epididymal white adipose tissue (eWAT) of adipocyte-specific LTA4H overexpression mice.

ABSTRACT: To investigate the alteration of eWAT gene expression in adipocyte-specific Lta4h overexpression mice(A-LTA4HOE mice), we collected the eWAT of control mice and A-LTA4HOE mice fed high-fat diet for 4 weeks or 16weeks after weaning at 4 weeks old. We performed gene expression profiling analysis using data from RNA-seq of these 4 groups.

The eWAT proteomics of wild type mice and Mdm2 adipocyte-specific knock-in mice fed high-fat diet

Project description:The epididymal adipose tissue (eWAT) samples from wild type mice fed high-fat diet for 12 weeks (H_WT_E) and Mdm2 adipocyte-specific knock-in mice fed high-fat diet for 12 weeks (H_KI_E) were mixed separately, and subjected to proteomic study by Label-free quantitative techniques and mass spectrometry-based proteomics techniques, etc. The proteomics of mixed eWAT samples were performed in Jingjie PTM BioLab (Hangzhou) Co. Ltd (www.ptm-biolab.com.cn). The difference was determined by 1.5-fold-change criterion, FDR < 0.01.

Project description:Purpose: The goals of this study are to identify GPCRs that are endogenously expressed by high fat diet fed mouse adipose tissue, we subjected RNA prepared from isolated mouse adipocytes (iWAT and eWAT) and BAT tissue to RNA-seq analysis. Methods: Total RNA extracted from mature adipocytes (iWAT and eWAT) and BAT tissue of C57BL/6 mice maintained on high fat diet for 12 weeks were used to construct high throughput sequencing libraries. RNAs with RIN >8 (assessed by the Agilent 2100 Bioanalyzer system) were used to prepare transcriptome libraries using the NEBNext Ultra RNA library prep kit (New England Biolabs). High throughput RNA-sequencing was performed using a Illumina HiSeq 2500 at the NIDDK Genomic Core Facility (NIH, Bethesda, MD). Raw reads were mapped to the mouse (mm9) genome. Results: Our study demonstrated that mouse adipocytes/BAT tissue from HFD-fed mice express several GPCRs.
