Project description:E0771.lmb tumors were grown after tamoxifen injection as previously described in wild-type and Shb iECKO (Shb flox/flox; Cdh5-CreERt2) mice, alternatively Shb ICKO (Shb flox/flox; Vav1-cre) mice. Mice were sacrificed on day 14-16 after tumor cell injection. The tumors were digested, CD45+-cells isolated by MACS microbeads (Miltenyi, Bergich Gladbach, Germany) and subsequently CD31+-cells isolated by fluorescence-activated cell sorting (FACS) after staining for CD31. RNA from EC CD31+-cells were subjected to bulk RNAseq.

Adiponectin-cre FTO flox/flox and FTO flox/flox mouse white fat tissues

Project description:Single nucleotide polymorphisms in intron 1 of the fat mass and obesity-associated (FTO) gene were found to be associated with an increased risk of adult obesity. Enhanced FTO expression in mice leads to hyperphagia, increased fat mass, and higher body weight. Neuronal-specific FTOâ??deleted mice have an identical lean body weight phenotype to global FTO-deleted mice. The physiological role of adipose FTO in the homeostasis of energy regulation remains to be elucidated. We used microarrays to elucidate the metabolic pathways that are regulated by FTO in the white fat. FTO flox/flox and Adiponectin-cre FTO flox/flox (AFO) mice were fed with chow diet. White fat tissues from epididymal adipose pad were harvested under ad lib condition for RNA isolation. Three independent pools of FTO flox/flox and AFO mouse white fat RNA were included in the study.

Parathyroid tissue transcriptome over 24h of PTHcre;Bmal1 flox/flox mice and of Bmal1 flox/flox mice

Project description:The objective is to elucidate the role of the circadian clock in the parathyroid glands. We investigate the parathyroid transcriptome of wildtype mice (Bmal1 flox/flox) and of mice with parathyroid-specific excision of the circadian clock gene Bmal1 (PTHcre;Bmal1 flox/flox) harvested at 4 hour interval for 24 hours. Rhythmic genes were identified by JTK_CYCLE analysis and revealed 1455 rhythmic genes of the Bmal1 flox/flox and 1055 rhythmic genes of the PTHcre;Bmal1 flox/flox. We found global damepning of circadian clock genes with abolished rhythmicity of Cry1, Cry2, Clock, Npas2, Rorc, and Usp2. We used GSEA analysis to investigate differential expression at each timepount and found downregulation of genes involved in ATP synthesis in PTHcre;Bmal1 flox/flox mice at 4 consecutive timepoints during the active period of the mouse.

Project description:Malignant peripheral nerve sheath tumors (MPNSTs) are the leading cause of premature death for patients with Neurofibromatosis type 1 and no approved targeted therapies are available. Transformation from Nf1-null benign plexiform neurofibromas is driven by the loss of the Cdkn2a (Arf) locus. Here, genetically engineered mouse models with combined Nf1 flox/flox and Arf flox/flox alleles were used (crossed with Postn-Cre+ mice). Tissue from MPNSTs that form in the Nf1-/-;Arf-/- setting were used for mRNA sequencing and compared to benign plexiform neurofibroma tissue (Nf1-/- from Nf1 flox/flox; Postn-Cre+ mice, GSE213789) to identify transcriptome signatures from MPNST and compare them to benign plexiform neurofibroma.

Gene expression of microglia from wild type, Gba flox/flox; Nestin-Cre, and Gba flox/flox; Nestin-Cre; Mincle-/- mice

Project description:To investigate the contribution of Mincle in neuronopathic Gaucher disease, we established Gba flox/flox; Nestin-Cre mice and crossed them with Mincle deficient mice. RNA sequencing of microglia from these mice revealed Axl, a phaogocytic receptor was upregulated in Gba flox/flox; Nestin-Cre mice. In addition, Tnf was upregulated in Gba flox/flox; Nestin-Cre mice in a Mincle-dependent manner. Our further study elucidated that Mincle, Axl and TNF are involved in the pathology of Gaucher disease.
