Effect of carnitine treatment on muscle gene expression in mice modeled on a high-fat diet

ABSTRACT: Skeletal muscle fatty infiltration has been shown to be strongly associated with sarcopenia. In this study，we constructed a high-fat diet mouse model and treated model mice with carnitine.Gene expression in skeletal muscle of different groups of mice was analyzed by RNA sequencing.

Eicosapentaenoic and docosahexaenoic acid - enriched high fat diet ameliorates sarcopenia-associated changes in the skeletal muscle transcriptome of C57BL/6J mice

Project description:Effects of EPA/DHA enriched high fat diets (HFD) compared to HFD-Corn oil after 8 weeks intervention on age/obesity induced sarcopenia. Results provides novel information on the signalling pathways regulated by EPA/DHA enriched HFD in delaying the onset of sarcopenia in C57Bl/6J mice compared to HFD-Corn oil. Four mice representing each diet group on the basis of body weight, plasma triglyceride and plasma cholesterol levels were selected for RNA isolation and microarray analysis. Total RNA from skeletal muscle of these selected animals was purified using the RNeasyÂ® Plus Universal Mini kit (Qiagen Nordic, Sollentuna, Sweden) by following the manufacturer?s instruction carefully. The extracted RNA was quantified by NanoDrop 2000c UV-Vis Spectrophotometer (Thermo Scientific, USA) and the quality of the RNA was estimated by RNA 6000 Nano LabChip for Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, California, USA).

Luteolin protects against obese sarcopenia in mice with high-fat diet-induced obesity by ameliorating inflammation and protein degradation in muscle

Project description:Although there are studies on obesity and sarcopenia, our understanding of obesity-mediated sarcopenia is insufficient. Additionally, no studies have investigated the application of luteolin in sarcopenia. We attempted to determine the effect of luteolin on obese sarcopenia in mice with high-fat diet (HFD)-induced obesity. Luteolin exerted suppressive effects on obesity, inflammation, and protein degradation in HFD-fed obese mice. Luteolin inhibited lipid infiltration into the muscle and decreased p38 and JNK activity and mRNA expression of inflammatory factors, such as TNFα, Tlr2, Tlr4, MCP1, and MMP2, in the muscle. Suppression of muscle inflammation by luteolin led to the inhibition of myostatin, FoxO, atrogin, and MuRF expression, which reduced protein degradation and improved muscle function. This is the first study to demonstrate that luteolin exerts a protective effect against obese sarcopenia, as its anti-obesity and anti-inflammatory activities inhibited protein degradation. Therefore, luteolin may be a useful supplement for prevention of obese sarcopenia.

Microarray analysis of skeletal muscle in aged mice

Project description:Sarcopenia is the age-induced, progressive loss of skeletal muscle mass and function, which is accompanied by reduced muscle performance. Individuals with sarcopenia often become bedridden or dependent on a wheelchair, leading to decreased quality of life. In this study, to better understand changes in skeletal muscle during sarcopenia, we performed a microarray analysis of skeletal muscle in young (13-week-old) and aged (26-month-old) mice. The microarray data shows that expression of the enzymes related to glucose and polyamine metabolism were decreased in aged mice compared with young mice.

P38 signaling underlies a cell-autonomous loss of stem cell self-renewal in aged muscle

Project description:Skeletal muscle aging results in a gradual loss of skeletal muscle mass, skeletal muscle function and decreased regenerative capacity, which can lead to sarcopenia and increased mortality. While the mechanisms underlying sarcopenia remain unclear, the skeletal muscle stem cell, or satellite cell, is required for muscle regeneration. Therefore, identification of signaling pathways affecting satellite cell function during aging may provide insights into therapeutic targets for combating sarcopenia. Here, we show that a cell-autonomous loss in self-renewal occurs via novel alterations in FGF and p38αβ MAPK signaling in old satellite cells. We further demonstrate that pharmacological manipulation of these pathways can ameliorate age-associated self-renewal defects. Thus, our data highlight an age-associated deregulation of a satellite cell homeostatic network and reveals potential therapeutic opportunities for the treatment of progressive muscle wasting. Satellite cells were isolated from young (3-6mo) and aged (20-25mo) adult mice; individual date files represent 2 independent pools of RNA from 4-8 mice at each timepoint.

Project description:In the current situation of the aging of the global population, primary sarcopenia is gradually attracting attention because of its potential risk to the elderly, high occult incidence and serious prognosis. As one of the most important hormones affecting skeletal muscle, the aging changes of thyroid hormone have been proved to be closely related to sarcopenia. Thyroid hormone exerts its effects locally in skeletal muscle by binding to thyroid hormone receptor. TRα is the main distribution type of thyroid hormone receptor in skeletal muscle. We have previously found the potential role of TRα in the aging process of skeletal muscle. As a nuclear transcription factor, TRα exerts subsequent biological effects by targeting regulatory genes. However, in the process of skeletal muscle aging, the target regulation map of TRα is still unclear.In this study, we constructed natural aging mouse models of different ages, combined ChIP-seq and mRNA-seq to explore the aging changes of TRα targets in skeletal muscle.
