Project description:Because the vagus nerve is implicated in control of inflammation, we investigated if brain death causes impairment of the parasympathetic nervous system, hence contributing to inflammation. Brain death (BD) was induced in rats. Anaesthetised ventilated rats (NBD) served as control. Heart rate variability (HRV) was assessed by ECG. The vagus nerve was electrically stimulated (BD+STIM) during BD. Intestine, kidney, heart and liver were harvested after 6h. Affymetrix chip- analysis was performed on intestinal RNA. Quantitative PCR was performed on all organs. Serum was collected to assess TNFα concentrations. Renal transplantations were performed to address the influence of vagus nerve stimulation on graft outcome. HRV was significantly lower in BD animals. Vagus nerve stimulation inhibited the increase in serum TNFα concentrations and resulted in down-regulation of a multiplicity of pro-inflammatory genes in intestinal tissue. In renal tissue vagal stimulation significantly decreased the expression of E-selectin, IL1β and ITGA6. Renal function was significantly better in recipients that received a graft from a BD+STIM donor. Our study demonstrates impairment of the parasympathetic nervous system during BD and inhibition of serum TNFα through vagal stimulation. Vagus nerve stimulation variably affected gene expression in donor organs and improved renal function in recipients. Project description:In France, new cancer cases keep on increasing with around 150 000 deaths yearly. Cancer therapy research is constantly evolving. Indeed, several studies explore new treatments or their combination with conventional cancer treatments. But, at the same time, complementary and alternative medicines, as osteopathy, remain little explored upon their role in the combination with conventional therapy.
Several studies showed indirect interaction between vagus nerve and cancer. Firstly, vagus nerve regulates homeostasis and immunity by reducing systemic inflammation while maintaining local inflammation and antitumor effects. Secondly, vagus nerve stimulation increases Heart Rate Variability (HRV). Moreover, a higher HRV is associated with an improvement of vital prognosis in cancer patients. Vagus nerve could be stimulated by noninvasive osteopathic manipulations.
This prospective, monocentric and randomized study is a collaboration between the Centre Hospitalier d’Avignon and the Institut de Formation en Ostéopathie du Grand Avignon. It focuses on using noninvasive osteopathic mobilizations to stimulate vagus nerve. Indeed, this study aims to evaluate effects of vagus nerve osteopathic stimulations on HRV in patients with lung cancer, colorectal cancer, Non Hodgkin Lymphoma or Multiple Myeloma. More specifically, this study will tell us whether vagus nerve noninvasive osteopathic stimulations induce increase of HRV associated with a decrease of systemic inflammation and an improvement of patient’s quality of life.

Methods: Blood was collected from Control and Treated patients followed by isolation of PBMC'S. Total RNA was extracted and RNA sequencing was done following standard protocols Results: Detailed analysis of the transcriptomic data show that anti-inflammatory responses were upregulated in the treated samples. Conclusions: This study showed the protective role of vagus nerve stimulation in epilepsy patients.

Project description:Guided by gut sensory cues, humans and animals prefer nutritive sugars over non-caloric sweeteners. But how the gut differentiates such stimuli to rapidly guide preferences remains unknown. In the intestine, innervated enteroendocrine cells synapse with the vagus nerve to convey luminal sugar stimuli to the brain within seconds. Here, we sequenced individual vagal nodose neuron cells from the left and right nodose to assess their relative contribution to sugar signaling from the gut. The neurons lack expression of sugar receptors and transporters, but do express receptors for neurotransmitters and neuropeptides secreted by intestinal sensory epithelial cells.

Project description:Peripheral nerve repair and functional recovery depend on the rate of nerve regeneration and the quality of target reinnervation. It is important to fully understand the cellular and molecular basis underlying the specificity of peripheral nerve regeneration, which means the achieving of respective correct pathfinding and accurate target reinnervation for regrowing motor and sensory axons. In this study, a quantitative proteomic technique, based on isobaric tags for relative and absolute quantitation (iTRAQ) was used to profile the protein expression pattern between single motor and sensory nerves at 14 days after peripheral nerve transection. Among a total of 1259 proteins identified, 176 proteins showed the differential expressions between injured motor and sensory nerves. Quantitative real-time RT-PCR and Western blot analysis were applied to validate the proteomic data on representative differentially expressed proteins. Functional categorization indicated that differentially expressed proteins were linked to a diverse array of molecular functions, including axonogenesis, response to axon injury, tissue remodeling, axon ensheathment, cell proliferation and adhesion, vesicle-mediated transport, response to oxidative stress, internal signal cascade, and macromolecular complex assembly, which might play an essential role in peripheral motor and sensory nerve regeneration. Overall, we hope that the proteomic database obtained in this study could serve as a solid foundation for the comprehensive investigation of differentially expressed proteins between injured motor and sensory nerves and for the mechanism elucidation of the specificity of peripheral nerve regeneration.

Project description:In this study, a quantitative proteomic technique based on isobaric tags for relative and absolute quantitation (iTRAQ) was used to compare the proteome of cultured sensory and motor nerve fibroblasts (Fbs). Among a total of 2597 overlapping proteins identified, we obtained 148 differentially expressed proteins, of which 116 proteins were significantly higher expressed in sensory Fbs, and 32 proteins were significantly higher expressed in motor Fbs. Western blot and qPCR analysis were applied to validate differentially expressed proteins. Functional categorization indicated that differentially expressed proteins were linked to a diverse array of biological processes , including regeneration, axon guidance, cytoskeleton organization, cell proliferation, cell migration, cell adhesion, and tissue remodeling, which might play a critical role in the specificity of peripheral nerve regeneration. Furthermore, following co-culture of motor neurons with motor or sensory Fbs , motor Fbs significantly enhanced neurite growth than sensory Fbs. These findings indicated that nerve Fbs expressed the distinct motor and sensory phenotypes involved in different patterns of protein expression, biological processes, and effects on neurons.
