Transcriptomic analyses of normal human pancreatic acinar cells

ABSTRACT: A cohort of 69 normal human pancreatic acinar cell specimens was collected from islet transplantation centers. Total RNA was isolated from uncultured pancreatic acinar cells, and sequencing was performed using Illumina NGS platforms. The resulting transcriptome data for each specimen were analyzed.

Epigenetic reprogramming in pancreatic acinar cell in the absence of MIST1

Project description:Epigenetic profile of tissues is reprogrammed under diseased conditions. H3K4Me3 and H3K27Me3 represent active and repressive epigenetic marks, respectively. ChIP-seq is an effective tool to study global protein-DNA interactions. To study global epigenetic differences, we used H3K4Me3 antibody to evaluate its enrichment in pancreatic acinar cells of WT and Mist1-/- mice followed by next generation sequencing. We found specific hotspots that showed differential enrichment for H3K4Me3 both in WT and Mist1-/- mice. The data show that pancreatic acinar cells are epigenetically reprogrammed under stressed cellular conditions. Global H3K4Me3 profiling of WT and Mist1-/- pancreatic acinar cells using ChIP-seq.

RNA-seq of purified human pancreatic dedifferentiated acinar cells and duct cells

Project description:Pancreatic acinar cells can dedifferentiate upon tissue injury and acquire ductal characteristics. This acquisition of duct cell features is critical in tumor development. Nevertheless, duct cells themselves are less prone for development of PDAC (pancreatic ductal adenocarcinoma) than dedifferentiated acini. We aimed to clarify which genes are unique for dedifferentiated acini. Mixed exocrine preparations of acinar and duct cells were obtained from human pancreatic donor organs and cultured to induce dedifferentiation. We lineage-labeled and FACS-purified these human dedifferentiated acinar cells and compared them to duct cells from the same donor (n=5).

Dynamic changes in the epigenetic landscape are mandatory for acinar cell reprogramming and pancreatic carcinogenesis [microarray]

Project description:Pancreatic acinar cell reprogramming is described as a common hallmark in pancreatic regeneration and carcinogenesis, although the regulatory mechanisms are barely understood. To overcome limitations of cellular diversity posed by heterogeneous cell compositions and high stroma density in pathogenic tissue specimens, we have established a pure in vitro system mimicking the pancreatic carcinogenic sequence. With this model, we globally profiled for epigenetic and transcriptional alterations and demonstrate that regulatory key players and structural genes are extensively regulated by epigenetic mechanisms. A conditional knockout of the epigenetic repressor Ring1b showed that particularly the epigenetic silencing of acinar differentiation genes is a crucial step in acinar cell reprogramming and pancreatic cancer development. Moreover, a persistent epigenetic repression of these genes manifests tumor cell malignancy. Depletion or drug-dependent inhibition of Ring1b promoted tumor cell reprogramming towards a less aggressive phenotype offering new options for therapeutic intervention. Simone,Benitz

PHARMACOLOGICAL INHIBITION AND REVERSAL OF PANCREATIC ACINAR DUCTAL METAPLASIA

Project description:Acinar ductal metaplasia (ADM), is believed to be one of the earliest precursor lesions towards the development of pancreatic ductal adenocarcinoma, and maintaining the pancreatic acinar cell phenotype suppresses tumor formation. We report that pStat3 and HDAC inhibition can attenuate ADM in vitro and TSA treatment reverses the dedifferentiated phenotype to one that is more acinar. Our findings suggest that pharmacological inhibition or reversal of pancreatic ADM represents a potential therapeutic strategy for blocking ductal reprogramming of acinar cells.

Gene expression of non-apoptotic pancreatic acinar cell death with autophagy

Project description:Serine protease inhibitor Kazal type 3 (Spink3) is a trypsin inhibitor in the pancreas. Spink3-/- pancreatic acinar cells are dead with excessive autophagy at birth (p0.5). To prove the role of nonapoptotic cell death with autophagy, we generated by transgenic technology the pancreas of Spink3-/-;XKI/+ mice contained both normal and dying acinar cells with autophagy. In this new mouse model, chronic inflammation occurred in the pancreas, indicating that some signals from nonapoptotic dead cell induce chronic inflammation in the pancreas. All samples were the pancreas at p0.5. Sample 1 and 2 are the pancreas from wild type (Spink3+/+, control) mice. Sample 3 and 4 are the pancreas from Spink3-/-, which all pancreatic acinar cells show induced nonapoptotic cell death with autophagy. Sample 5 and 6 are the pancreas from Spink3-/-XKI/+, about half acinar cells are normal, but other acinar cells show induced nonapoptotic cell death with autophagy.

RBPJ deficiency sensitizes pancreatic acinar cells to Kras-mediated PanIN initiation

Project description:Pancreatic ductal adenocarcinoma (PDAC) has a poor prognosis and is among the most common causes of cancer death worldwide. KrasG12D mutation is the main driver mutation but insuffi-cient for progression of invasive cancer on its own indicating that other pathways, such as Notch signaling may participate in that process. RBPJ, the only transcription factor in Notch signaling, is frequently lost in human cancers and associated with more aggressive breast cancer phenotype. Notch-independently, in complex with P48, RBPJ initiates transcription of its paralogue RBPJL ensuring acinar differentiation. The RBPJ knockout has an embryonic lethal effect. Mice with pancreas-specific deletion of RBPJ presented with less acinar tissue and large duct-like structures suggesting a relevance of RBPJ for acinar to ductal reprogramming and even pancreatic neoplasia. Here, we found reduced RBPJ expression level in the human PDAC specimens. Analyses of transgenic mouse models of an inducible P48-dependent RBPJ knockout revealed that it is dispensable for the maintenance adult acinar cells. In the context of oncogenic KRAS expression, the RBPJ deficiency facilitated the development of PanIN lesions with massive fibrotic stroma. Interestingly, RNA seq data revealed corresponding transcription pattern prior to phenotypic alterations.
