Transcriptome profiling of HCT116 cells and differentiated HCT116 cells induced by forced expresssion of MYOD1 or PPARG using RNA-sequencing.

ABSTRACT: The core proeprty of cancer cells is neural stemness, which determines tumorigenicity and pluripotency. Therefore, cancer cells can supposedly be induced to differentiate along different lineages. HCT116 is a human colon cancer cell line. We found that HCT116 cells differentiated into muscle- or adipocyte-like cells upon induction by muscle or adipocyte differentiation factor MYOD1 or PPARG, respectively.

Therefore, cancer cells can supposedly be induced to differentiate along different lineages. B16F10 is a mouse melanoma cell line. We found that B16F10 cells differentiated into muscle cells or adipocytes upon induction by muscle or adipocyte differentiation factor Myod1 or Pparg, respectively. Moreover, Setdb1 is an epigenetic modification factor responsible for catalyzing the histone modification H3K9me3. Transcription of Setdb1 gene is enriched in embryonic neural cells during vertebrate embryogenesis. Setdb1 is upregulated in cancer cells and promotes cancers. We found that knockdown of Setdb1 in B16F10 cells led to a neuronal-like differentiation phenotype.

Project description:The core proeprty of cancer cells is neural stemness, which determines tumorigenicity and pluripotency. It has been reported that neural stem cells are uniquely immune-privileged, suggesting that neural stemness also confers cancer cells with the ability of immune evasion or immune privilege. CT26 is a mouse colon cancer cell line. We found that dedifferentiation of CT26 cells with overexpression of Oct4 and SOX2 enhanced neural stemness and decreased immunogenicity in cells.

ncRNA DUBR RAP-MS in HCT116 cells

Project description:Non-coding RNAs (ncRNAs) are finely tuned cellular regulators important for human cell growth and cancer progression. DUBR (also known as linc00883) is a nuclear ncRNA first discovered in mice for its role in regulating myoblast differentiation through interactions with chromatin and DNA methyltransferases. High expression levels of this ncRNA are predictive of poor patient outcome in colon adenocarcinoma, suggesting that DUBR may be involved in controlling cancer growth. To elucidate its function, we used RAP-MS and RNA immunoprecipitation techniques which revealed its interaction with epigenetic maintenance proteins in the human colon cancer cell line HCT116. Further, ATAC-seq and RNA-seq were used to address its function in regulating the epigenome and transcriptome of HCT116 cells. Here we report that DUBR is a regulator of human colon cancer cell line HCT116 survival.
