The gut microbiota regulates the vascular inflammatory phenotype in mesenteric ischemia-reperfusion injury via endothelial Toll-like receptor-2 signaling

ABSTRACT: Here, we set out to study the impact of gut commensals and TLR2 signaling on the vascular inflammatory phenotype of acute mesenteric I/R injured venules. Taking advantage of gnotobiotic experimentation and an endothelial-specific Tlr2-deficient mouse model, we dissect whether I/R-induced NETosis is also supported by endothelial TLR2 activation. To resolve the engagement of microbiota-induced TLR2 signaling on neutrophil function, based on agonist-treatment, we pinpoint the role of TLR2/1 vs TLR2/6 signaling in ex vivo NETosis induction.
