PTEN restrains tumor expansion through cell autonomous and non-autonomous mechanisms

ABSTRACT: To gain insight into transcriptional changes that occur in SmoM2 SHH-MB tumor cells that lack Pten and could account for the cell autonomous changes we observed in tumors of a mouse model, we performed single cell RNA-sequencing (scRNA-seq) of whole tumors dissected from P22 SmoM2 (Atoh1-FlpoER/+; R26MASTR/LSL-SmoM2) and P16 SmoM2-Ptenfl/fl (Atoh1-FlpoER/+; R26MASTR/LSL-SmoM2; Ptenfl/fl) mice (n = 2 per genotype).

Project description:Ptk2 was identified as a targetable genetic vulnerability in glioblastoma (GBM) cells. Our objective was to test the effect of Ptk2 deletion on survival and tumor phenotype in a mouse model of GBM. At P0, mice carrying Trp53fl/fl (Trp53tm1Brn, JAX #008462), Ptenfl/fl (Ptentm1Hwu/J, JAX #006440), Rb1fl/fl and R26-LSL-Cas9-GFP (JAX #026175) alleles were subjected to stereotactic injection to deliver lentiviral particles expressing Cre and either Ptk2-targeting or control gRNAs to the NSCs residing in the lateral subventricular zone. This injection initiates a GBM tumor in the mice. At time of sacrifice, tumor tissue was harvested and RNA was extracted for sequencing.

RNA sequencing of genetically engineered mouse model lung tumors and normal mouse lung.

Project description:Genetically engineered mouse models (GEMM) of cancer are powerful tools to study multiple aspects of caner biology. We developed a novel GEMM for lung squamous cell carcinoma (LSCC) by genetically combining overexpression of Sox2 with loss of Lkb1: Rosa26LSL-Sox2-IRES-GFP;Lkb1fl/fl (SL). We compared gene expression profiles of SL lung tumors with normal mouse lung tissue, mouse lung adenocarcinoma (LADC) tumors from KrasLSL-G12D/+;Trp53fl/fl (KP), mouse LSCC tumors from Lkb1fl/fl;Ptenfl/fl (LP) model as well as Lenti-Sox2-Cre Lkb1fl/fl.

SOX2 genomic binding sites in lung squamous cell carcinoma GEMM tumor and cell lines

Project description:SOX2 is a lineage specifier oncogene for lung squamous cell carcinoma (LSCC) and frequently amplified and overexpressed in human LSCC tumors (up to 90% of the cases). Our study demonstrated that SOX2 is a key determinant of neutrophil recruitment to tumors even in the absence of squamous histology. We generated cell lines from KrasLSL-G12D/+;Trp53fl/fl (KP) tumors that overexpress Sox2 (i.e. tumors from Lenti-Sox2-Cre infected KP mice that are validated to have Sox2 overexpression) (abbreviated as KPS) and employed chromatin immunoprecipitation sequencing (ChIP-seq) to identify genomic binding loci of SOX2 in KPS lines as well as Lkb1fl/fl;Ptenfl/fl (LP) LSCC tumors.

Project description:We report scRNA-seq data showing the effect of Prkaa1/2 deletion on medulloblastomas that form in Smo-mutant mice. We generate tumor-bearing mice by breeding hGFAP-Cre/Prkaa1fl/fl/Prk112fl/fl and Prkaa1fl/fl/Prk112fl/fl/SmoM2 mouse lines to prodce tumor-prone progeny with homozygous, conditional deletion of Prkaa1 and Prkaa2 (G-SmoAMPK mice). We compared tumors from G-Smo mice withintact Prkaa1 and Prkaa2 to tumors from G-SmoAMPK mice, using Drop-seq-based scRNA-seq.

Project description:We wished to investigate the role of E-cadherin loss in our mouse parietal cell/pre-parietal cell E-cadherin knock-out, p53 knock-out, oncogenic Kras induced model of gastric cancer. As such, we isolated RNA from stomach tissue from our E-cadherin knock-out model (Atp4b-Cre;Cdh1(fl/fl);Kras(LSL-G12D/+);Trp53(fl/fl);Rosa26(LSL-YFP/LSL-YFP)) and our E-cadherin heterozygous model (Atp4b-Cre;Cdh1(fl/+);Kras(LSL-G12D/+);Trp53(fl/fl);Rosa26(LSL-YFP/LSL-YFP)). We then performed a microarray on this stomach tissue from four independent mice of each genotype. Differentially expressed genes were identified and gene set overlap analysis was used to identify pathways enriched in one model over the other.
