The Role of astrocytic TCF7L2 in somatosensory cortex development and function

ABSTRACT: Astrocytes undergo extensive differentiation and maturation during postnatal development. While some mechanisms guiding astrocyte development are beginning to be identified, the molecular mechanisms that link astrocytic maturation with neuronal function remain incompletely understood. We investigated the role of the Wnt/β-catenin effector TCF7L2 transcription factor in postnatal astrocytes using single-nucleus RNA sequencing. We discovered that loss of astrocytic Tcf7l2 resulted in broad transcriptional dysregulation across astrocytes particularly in genes invovled in synaps development, function, maturation, governing amino acid, ion transport and membrane potential. These findings identify the novel role of Wnt/β-catenin effector TCF7L2 as a critical regulator of astrocyte-mediated neurophysiological support and underscore the importance of astrocytic maturation during a postnatal developmental window.
