ABSTRACT: To check the effectes of ST8Sia6 overexpression we performed total RNS sequesnning in porcine kidney endothelial cells to understand the biological functions of DEGs associated with ST8Sia6 overexpression in porcine kidney endothelial cells upon co-culture with human PBMCs, transcriptomic analysis was performed on four independent experimental conditions, including control GFP-expressing cells and ST8Sia6-overexpressing cells.

Project description:Endothelial cells of xenotransplanted gene-edited porcine kidneys are important due to the initial and prolonged exposure to host immune cells. To better study this cell population kidney endothelial cell lines (KECs) were generated by double CD31+ sorting. In order to ensure the endothelial identity of the cells RNA-seq was performed on KECs and the concomitant CD31- population from the initial sort. Analysis of gene expression in KECs showed a depletion of markers associated with tubular identity (SLC34A1, SLC2A2, SLC12A1), and enrichment for markers of endothelial (PECAM1, CDH5, VWF) and kidney endothelial (EHD3, HOX87, HOX89) identity.

Project description:Succesful xenotransplantation of porcine kidneys into humans requires the ubiqutous expression of certain human transgenes. Modern transcriptomics now alllows gene expression to be measured at the per-cell level, which can provide important insights into the expression pattern of these life-sustaining transgenes in a complex organ such as the kidney. Analysis of this scRNA-seq data shows ubiquitous expression of transgenes in most major kidney cell populations which includes various subtypes of endothelial cells.

Transcription profiling of S. cerevisiae response to nitrosative stress

Project description:Nitric oxide and NO-derived species (RNS) are defense molecules with broad antimicrobial activity. Micro-organisms have developed strategies to sense RNS and counteract their damaging effects. We used Saccharomyces cerevisiae, harbouring a deletion of YHB1 that encodes the main NO scavenger enzyme, to study consequences of RNS exposure on whole genome transcriptional response. The expression of >700 genes was altered on RNS treatment. No major role for ROS-scavenging enzymes was found, and the respiratory chain, the main site of ROS production, had only minor involvement in the RNS-induced stress. The changes were generally transient and also found after treatment with the respiratory inhibitor myxothiazol. 117 genes however showed a persistent response which was not observed after myxothiazol treatment. Of these, genes of the glutathione and DNA repair systems, iron homeostasis and transport were found up-regulated. Severe repression of genes of respiratory chain enzymes was observed. Many of these genes are known to be regulated by the transcription factor Hap1p suggesting that RNS might interfere with Hap1p activity. We showed also that Msn2/4p and Yap1p, key regulators of the response to, respectively, general stress and oxidative stress, played a role in mediating the RNS-induced response.
