Effect of iPS cell culture medium on the maturity of induced cardiac organoids

ABSTRACT: The quality of iPSCs was compared using two types of culture media. PluriTest scores were 27.4 ± 1.1 for iMatrix511+StemFit and 35.2 ± 0.5 for LN521+mTeSR. n = 3. LN521+mTeSR was significantly higher than iMatrix511+StemFit. The quality of iPSCs was compared using two types of culture media. KaryoStat assays revealed no genetic or karyotypic mutations in both iMatrix511+StemFit and LN521+mTeSR. n = 3.

Project description:In the following experiment, three different hESC cell lines (HES2, MEL1 and H9) were grown in the presence of KOSR, KOSR or mTESR containing media respectively. KOSR (Knockout serum replacement medium) is a standard media allowing the growth of hESC without the need for manual passaging - Enzymatic passaging is used instread. mTESR (Ludwig et al., 2007) is a media allowing the growth of hESC on matrigel with enzymatic passaging. At day 7 after passaging, these cells were FACs sorted for the presence of GCTM-2 and CD9 into 4 distinct fractions (p4: GCTM-2-neg, CD9-neg; p5: GCTM-2-low, CD9-low; p6: GCTM-2-medium, CD9-medium and p7: GCTM-2-high, CD9-high). For each cell line-subfraction combination, RNA was harvested and subject to microarray. From each experiment (individual cell line), 4 samples were collected (p4, p5, p6 and p7) and these were subject to microarray. In each case the experiment was performed in triplicate

Transcription profiling of human karyotypically different embryonic stem cells

Project description:Recent reports indicated that during long term culture HESCs acquired cytogenetic abnormality which are similar to those occurred in tumorigenesis. This raise concerns about the safety use of cultured HESCs. The cause of genetical instability in cultured HESCs is poorly understood and whether these changes will progressively developed and ultimately transform HESCs into malignancy has not been systematically investigated. In our lab, chHES3 was found to have progressive chromosomal changes during long term suboptimal culture. In order to understand the molecular mechanism underlying the karyotypic changes, we compared the expression profile of differnt karyotypic chHES-3 cells and signa pathways involved in the process. We found that the expression profiles of oncogenes, suppress tumor genes and genes related with apoptosis had tendency to EC with complexity and compared with the normal karyotypic HES cells, the activating Wnt pathway gene up-regulated and the suppressing gene down-regulated in the aberrant karyotypic HES cells. The species and abundance of the wnt gene family had obvious difference in difference batch mouse embryonic fibroblast feeder cells. We used microarrays to detail the global programme of gene expression underlying cellular mailignant transformation, identified distinct genes or gene clusters and analysed some signal pathways involved in this process. Experiment Overall Design: Different karotypic human embryonic stem cells and human embryoinc carcinoma cells were collected for RNA extraction and hybridization on Affymetrix microarrays. By the karyotype analysis we selected three different karyotypic chHES-3 cells and human embryonic carcinoma cells to compared.

Establishment of Pig Embryonic Stem Cells using Simplified Serum-free media and Adaptation of Feeder-free conditions

Project description:Most porcine embryonic stem cells (pESC) derived from in vitro blastocysts Still require complex media compositions and feeder layers which make difficulty to adapt their broad and routine use. We derived pESC using simplified culture media (FIW) consisting of small molecules FGF2, IWR-1, and WH-4-023 in serum-free medium. We established several pESC-FIW lines capable of single cell passaging with short cell doubling time (about 12 hours). These cells expressed pluripotency markers, OCT4, SOX2, and NANOG, as well as cell surface markers, SSEA1, SSEA4, and TRA-1-60. pESC-FIW showed stable proliferation rate and normal karyotype even after long-term culture over 50 passages. Established pESC showed formative characteristics based on the negative expression for the naive marker KLF4 and primed marker T, whereas strong expression for the formative marker OTX2 with high levels of related genes. Transcriptome analysis showed that pESC-FIW had similar cellular identities to reported pESC maintained in complex media formulations and had characteristics of gastrulating epiblast cells. In addition, pESC-FIW could be maintained for multiple passages in a feeder-free condition on fibronectin coated plate using mTeSR™, a commercial medium for feeder-free culture. These results indicate that the canonical WNT (IWR-1) inhibition and the SRC inhibition (WH-4-023) are sufficient to establish pESC capable of single cell passaging and feeder-free expansion under the serum-free condition. Easy to maintain-pESC can be a tool for gene editing application useful for agriculture and biomedicine, and lineage commitment studies.

Feeder-Independent Derivation of Induced-Pluripotent Stem Cells From Peripheral Blood Endothelial Progenitor Cells

Project description:The objective of this study was to reprogram peripheral blood-derived late-endothelial progenitor cells (EPCs) to a pluripotent state under feeder-free and defined culture conditions. Late-EPCs were retrovirally-transduced with OCT4, SOX2, KLF4, c-MYC, and iPSC colonies were derived in feeder-free and defined media conditions. EPC-iPSCs expressed pluripotent markers, were capable of differentiating to cells from all three germ-layers, and retained a normal karyotype. Transcriptome analyses demonstrated that EPC-iPSCs exhibit a global gene expression profile similar to human embryonic stem cells (hESCs). We have generated iPSCs from late-EPCs under feeder-free conditions. Thus, peripheral blood-derived late-outgrowth EPCs represent an alternative cell source for generating iPSCs.

PEDF was detected abundantly in culture supernatant media of primary and iPSC-derived RPE. We examined the gene expression in primary RPE and iPS-derived RPE.
