Gene expression profile on pons and medulla of maternal immune activation model rat

ABSTRACT: We created two-hit immune activation model by inducing a pseudo-bacterial infection (administration of lipopolysaccharide (LPS)) on postnatal (P) 11 in pups born to rats infected with a pseudo-virus (administration of polyinosinic-polycytidilic acid (poly I:C)) during pregnancy of day 10. Our model rats showed increased mortality rate due to LPS administration on P11 and abnormal blood biochemical test results. Next, we examined the gene expression in pons and medulla oblongata of our model rats on P11 by DNA microarray and found that a number of genes showed more than a 2-fold change in expression.

Project description:Identify differentially expressed genes related to the neurodegenerative process in a new animal model of hepatic encephalopathy (HE). The animal model consists on the simulation of several bouts of HE in PCA rats, being the precipitant factors of the episodes ammonia (NH3) and/or lipopolisaccharide (LPS) or saline. Regular administration (one every two weeks up to 10 infussion) of NH3 (20 μl/min along 3h) and/or LPS (5mg/kg) or in PCA rats. Sham rats were used as a surgery control.

Spatial transcriptomics in the postmortem brains of severe acute COVID19 [Pons]

Project description:Understanding the pathological basis of the neurological symptoms observed following SARS-CoV2 infection is essential to optimizing outcomes and developing therapeutics. To date, small sample sizes and narrow molecular profiling limit the generalizability of findings. We profiled multiple cortical and subcortical regions in postmortem COVID-19 patient brains and controls (total n=42) with spatial transcriptomics in the frontal cortex, midbrain and pons, bulk RNA expression in frontal cortex, midbrain, basal ganglia, and pons, and proteomics in the 8 regions (frontal cortex, temporal cortex, occipital cortex, midbrain, pons, hippocampus, thalamus and basal ganglia). We observed a multi-regional antiviral response in the absence of direct active SARS-CoV2 infection. We identified dysregulation of mitochondrial and synaptic pathways in deep-layer excitatory neurons and up-regulation of neuroinflammation in glia, in both mRNA and protein. Remarkably, these alterations overlapped substantially with changes in various age-related neurodegenerative diseases, such as Parkinson’s disease (PD) and Alzheimer’s disease (AD). Our work, combining multiple experimental and analytical methods, demonstrates the brain-wide impact of severe COVID-19, involving both cortical and subcortical regions, shedding light on potential therapeutic targets within pathways typically associated with pathological aging. Gene expression profiling of cultured cells from brainstem of spontaneously hypertensive and Wistar Kyoto rats

Project description:Hypertension is a multifactor disease that possibly involves alterations in gene expression in hypertensive relative to normotensive subjects that are largely unknown. In this study we used high-density oligoarrays to compare gene expression profiles in cultured neurons and glia from pons and medulla oblongata of newborn spontaneously hypertensive (SHR) and normotensive Wistar Kyoto (WKY) rats, a widely documented animal model of hypertension. We found 358 genes differentially expressed between SHR and WKY brainstem cells that preferentially map to 24 metabolic/signaling pathways. Some of the pathways and regulated genes identified herein are obviously related to blood pressure regulation; however there are several genes differentially expressed in SHR not yet associated to hypertension or participating in blood pressure regulation. These constitute a rich resource for the identification and characterization of novel genes involved in hypertension development, or associated to phenotypical differences observed in SHR relative to WKI. In conclusion, this study describes for the first time the gene profiling pattern of brainstem cells from SHR and WKY rats, which opens up new possibilities and strategies of investigation and possible therapeutics to hypertension, as well as for the understanding of the brain contribution in this pathology. Gender differences in the cytokines and chemokines gene expression profiles within the medulla oblongata of SHRs

Project description:Medulla oblongata is a region of the brain containing multiple blood pressure regulation centers, including the nucleus of the solitary tract (NTS), a pivotal region for regulating the set-point of arterial pressure. Our previous findings suggest that the NTS of male pre-hypertensive rats and spontaneously hypertensive rats (SHRs) exhibits abnormal inflammatory condition compared with that of normotensive Wistar-Kyoto (WKY) rats. Females of SHRs exhibit a lower blood pressure than their male counterpart but the molecular mechanism is not well described. To unveil the gender differences in the central regulation of blood pressure, we investigated the gene expression profile of cytokines and chemokines in the medulla oblongata of female SHRs compared to that of their male counterparts.
