Transcriptomic analysis of pancreas from Elp3 epithelial-specific knockout and wild-type mice with and without caerulein treatment

ABSTRACT: To study the role of Elp3 in pancreatic homeostasis and pancreatitis, pancreas tissues were collected from Elp3(fl/fl); Pdx-Cre and wild-type mice either untreated or after caerulein-induced pancreatitis. RNA sequencing was performed to identify transcriptional changes associated with Elp3 loss and pancreatic injury.

mRNA expression pattern of KrasG12D mice with acute pancreatitis compared with wild-type mice without acute pancreatitis

Project description:Transgenic KrasG12D mice can recapitulate pancreas intra-epithelial neoplasia (PanIN). Caerulein is a cholecystokinin analogue and induces acute pancreatitis when injected intra-abdominally. Caerulein-induced acute pancreatitis will accelerate PanIN progression in KrasG12D mice. We compared mRNA profile changes between KrasG12D mice with acute caerulein-induced pancreatitis and wild-type mice without acute pancreatitis. The experiment had two groups. Experiment group: KrasG12D mice with acute caerulein-induced pancreatitis (N=6). Three mice in experiment group received 1-week caerulein injection, and the other three mice received 2-week caerulein injection. All experiment group mice started to receive caerulein injection at 1-month of age, and were sacrificed at the last day of caerulein injection. Control group: wild-type mice without acute pancreatitis (N=6). The mice were sacrificed at 1.5-month of age. Whole pancreas tissue lysate samples were subjected to mRNA array assay.

Single-cell transcriptomic analysis of mouse pancreas: characteristic features of pancreatic ductal cells in chronic pancreatitis

Project description:Chronic pancreatitis (CP) is a pathogenically complex fibro-inflammatory disorder of the pancreas. Our understanding of CP pathogenesis is partly limited by the incomplete characterization of pancreatic cell types. Here, we performed single-cell RNA sequencing on 3,386 cells from the pancreas of one control mouse and mice with caerulein-induced CP. These data provides a preliminary description of the single-cell transcriptome profiles of mouse pancreata and accurately demonstrates the characteristics of pancreatic ductal cells in CP.

Expression data of caerulein-treated wild-type mice and KrasG12D-mutated mice at different stages of pancreatic regeneration after inflammatory injury

Project description:Consecutive caerulein injections induce an acute pancreatitis in mice. Here, we recorded gene expression levels at different stages of pancreatic regeneration in wild-type mice as well as KrasG12D-mutated mice. Tissue was collected from mice pancreata, cell sorting was not performed. t=0h refers to the time where caerulein was injected. control referes to NaCl-treated samples (no caerulein). 13 time points were used for wild-type mice, 9 time points were used for KrasG12D-mutated mice. Multiple replicates were generated for each time point. We used Affymetrix GeneChip Mouse Gene 1.0 ST arrays.

Heparin-binding proteome in normal pancreas and murine experimental pancreatitis_C4PR_LIV

Project description:Acute pancreatitis (AP) is acute inflammation of the pancreas, mainly caused by gallstones and alcohol, driven by changes in communication between cells. Heparin-binding proteins (HBPs) play a central role in cell communication. Therefore, we used heparin affinity proteomics to identify extracellular HBPs in pancreas and plasma of normal mice and in a caerulein mouse model of AP. Many new extracellular HBPs (360) were discovered in the pancreas, taking the total number of HBPs known to 786. Extracellular pancreas HBPs form highly interconnected protein-protein interaction networks in both normal pancreas (NP) and AP. Thus, HBPs represent an important set of extracellular proteins with significant regulatory potential in the pancreas. HBPs in NP are associated with biological functions such as molecular transport and cellular movement that underlie pancreatic homeostasis. However, in AP HBPs are associated with additional processes such as acute phase response signalling, complement activation and mitochondrial dysfunction. Plasma HBPs in AP included known AP biomarkers such as serum amyloid A, as well as emerging targets such as histone H2A. Pancreas HBPs are extracellular and so easily accessible and are potential drug targets in AP, whereas plasma HBPs represent potential biomarkers for AP.
