Gene expression profile at single cell level of ID8 ovarian cancer chemotherapy model

ABSTRACT: Chemotherapy is the first-line treatment for ovarian cancer, yet chemoresistance frequently develops at the late stage of treatment. In this study, we established the preclinical model of metastatic, syngeneic high-grade serous ovarian cancer (HGSOC) model using the ovarian cancer cell line ID8 that recapitulates the full chemotherapy course. 4-5 weeks following intraperitoneal injection, tumour grows extensively in the abdomen of the mice, which were subsequentially treated with cisplatin. Early treatment cycles significantly suppressed tumor growth, including a marked response during the first and subsequent cycles following initial relapse. Nevertheless, all tumors ultimately relapsed with chemoresistant phenotypes after two treatment cycles , mirroring the clinical course often observed in the treatment of HGSOC patients. We conducted single-cell RNA sequencing (scRNA-seq) on both parallelly debulked primary tumors and their corresponding resistant counterparts to elucidate changes in cellular composition.
