Transcriptome profiling of senescent and non-senescent BPH epithelial cells after CD4⁺ T cell co-culture

ABSTRACT: Human primary CD4⁺ T cells were co-cultured with either senescent or non-senescent primary BPH epithelial cells for 6 hours. BPH epithelial cells were then isolated and subjected to RNA sequencing. The experiment was repeated three times independently.

Project description:BPH-1 prostate epithelial cells when cultured in Matrigel form 3D acini which closely recapitulate the tissue architecture of the prostate. The presence of primary stroma increases epithelial adhesions and causes changes in morphology. To identify the stromal signals which control actin/cadherin dynamics in epithelial cells we performed a microarray analysis on the stroma using Affymetrix U133 plus 2.0 arrays. Primary stroma from 7 different patients were cultured with (PSE) or without (PS) 3D BPH-1 epithelial acini to determine the gene expression changes within the stroma. The RNA from the Primary epithelia and stromal cultures were used at passages 1 or 1-3, respectively.

RB vs. SB: a microarray analysis of genes that were differentially expressed in resistant seedling pool and susceptible seedling pool both infested with BPH for 24h. RB vs. RN: a microarray analysis of genes that were differentially expressed in resistant seedling pool infested with BPH for 24h and resistant seedling pool without BPH infestation. Goal was to explore the molecular basis underlying BPH-resistance in rice.

Therapeutic potential of pomegranate juice-derived nanovesicles in nude mouse benign prostatic hyperplasia (BPH) xenograft model

Project description:Benign prostatic hyperplasia (BPH) and associated lower urinary tract symptoms affect a large percentage of the male population and places a substantial burden on the world health system. Current therapies include 5-alpha reductase inhibitors and alpha-blockers that are only partially effective and pose a huge economic burden, emphasizing the urgent need for effective, economical therapies. We isolated nanovesicles from pomegranate juice (Punica Granatum) (referred to as ‘POM-NVs’) and report to our knowledge for the first time, that these vesicles possess therapeutic potential against BPH. Following extensive characterization of POM-NVs, we tested their therapeutic potential in vitro using BPH1 cell line and identified a potential anti-proliferative and pro-apoptotic effect. We further tested these vesicles using a clinically relevant xenograft mouse BPH model derived from human BPH tissues. Remarkably, POM-NVs could reverse the BPH phenotype conferred by TGF-β mediated signaling and induced epithelial-to-mesenchymal (EMT) reversal, leading to the restoration of prostate epithelial states in vivo and in vitro. Furthermore, these vesicles attenuated bone morphogenic protein 5 (BMP5) signaling, a cardinal alteration that is instrumental in driving BPH. Considering the large incidences of BPH and its associated economic burdens, our study has important implications and can potentially improve the clinical management of BPH.

Transcriptional alterations in BPH

Project description:We identified a BPH transcriptional signature that included 392 significantly differential expressed genes between BPH and control samples, and validated this BPH transcriptional signature using two independent study cohorts. By performing integrative analysis using transcriptional and methylation profiling, we identified two distinct BPH subtypes, supporting robust biologically distinct subgroups across different data types. To validate these BPH subtypes, we tested our signature via k-means clustering in two independent cohorts, and identified nearly identical subgroups. To nominate potential subtype specific therapeutic options, we utilized a Connectivity Map analysis, and found 50% of nominated compounds in one subgroup were related to inhibition of mTOR signaling.
