Quantitative phosphoproteomic analysis reveals involvement of PD-1 in multiple T cell functions

Project description:The programmed cell death protein 1 (PD-1) pathway is a critical inhibitory checkpoint for T cells, and antibodies blocking PD-1 promote immune-mediated identification and clearance of malignant cells. Despite the success of these antibodies, the majority of patients do not respond to PD-1 blockade and many experience immune-related adverse events. As such, there is an urgent need to better understand PD-1 signaling, not just in order to explain the mechanism behind the unfavorable responses, but also to develop next-generation therapeutics. We performed global quantitative phosphoproteomic interrogation of PD-1 signaling in human T cells and by complementing our analysis with functional validation assays in primary human cells. To investigate the T cell phosphoproteome, Jurkat cells were treated with either a) anti human CD3 antibody, b) anti human CD3 antibody together with recombinant human PD-L2-Fc or c) IgG1 isotype control antibody for 30 seconds, 5 minutes or 15 minutes. The global and phosphoproteome was quantified using an isobaric labeling approach (e.g. TMT) in combination with TiO2 enrichment and three different pY specific antibodies.

We profiled (transcriptome analysis) effects of anti-PD-L1 antibody therapy on gene expression in B16 melanoma tumors of B cells depleted and WT syngeneic mice. After 7 days of B16 tumors implantation, mice were treated or untreated with anti-PD-L1 antibody (every three days).

Systemically administered HMGN1 immunostimulatory peptide with anti-PD-L1 antibody exerts synergistic antitumor effects

Project description:In this study, we explore a new combination therapy of anti-PD-L1 antibody with an immunostimulatory peptide derived from an alarmin high mobility group nucleosome binding protein 1 (HMGN1). Combination treatement of HMGN1 immunostimulatory peptide with anti-PD-L1 antibody exerted robust anti-tumor effects in Colon26 subtaneous murine model. To understand transcriptomic differences of immune cell population in the tumor-bearing mice after treated with single-agent or combination therapy of HMGN1 with anti-PD-L1 antibody, we performed transcriptome analysis on tumor tissue using 5’end Serial Analysis of Gene Expression (SAGE)-sequencing with an Ion 540 Chef kit, an Ion 540 Chip kit, and an Ion S5 Sequencer (Thermo Fisher Scientific).

The impact of PD-1 antibody or G-CSF antibody on the immune system and the hematopoiesis of TNBC mice

Project description:Triple negative breast cancer (TNBC) is the most intractable subtype of breast cancer. Inhibitors of programmed cell death protein-1 (PD-1) or its ligand PD-L1 have been considered as promising treatment for TNBC patients. However, only a minor subset of TNBC patients benefits from the monotherapy. An analysis of how PD-1/PD-L1 blockage affect immune activities in TNBC tumor-bearing mice could provide insights for it limitation. G-CSF was known to be highly elevated in TNBC cells, and affecting hematopoiesis and the immune system. Spleen is an immune organ and with extramedullary hematopoiesis in TNBC mice. Here we analyzed splenocyte gene expressions changed by administration of either PD-1 antibody or G-CSF antibody in both 4T1 tumor bearing mice and non-tumor control mice. TNBC tumor significantly changed gene expressions in splenocytes, which is consistent with a significant change in splenocyte composition. These genes were enriched in immune related pathways. Anti-PD-1 antibody treatment had limited impact on spleen immune cells composition and splenocyte gene expressions. The most affected genes were enriched in metabolism and extracellular matrix related pathways. Anti-G-CSF antibody treatment had higher impact on the splenocyte gene expression profile, making it more like the gene expression profile in the non-tumor control mice.

Effects of AKT/WEE1 inhibitors or ALDH inhibitor combined with PD-1 antibody on melanoma

Project description:The effects of AKT/WEE1 inhibitors or ALDH inhibitor combined with PD-1 antibody on melanoma were examined. Mouse B16F10 melanoma cells were subcutaneously injected into C57/B6 mice. When tumors were well-vascularized, mice were treated with anti-PD-1, AKT I (AZD5363)+WEE1 I (MK1775), AZD5363+MK1775+anti-PD-1, ALDH I (KS100), KS100+ anti-PD-1 daily. Mice were sacrificed when tumors in the control group (DMSO treated) reached 2000 mm3 and tumors were removed. RNA was extracted from tumor samples and gene expression was analized using RNAseq analysis.
