Placental Remodeling in Gestational Diabetes Mellitus (GDM) Disrupts Lipid Metabolism

Project description:Gestational diabetes mellitus (GDM) is a pregnancy-specific metabolic disorder associated with adverse maternal and fetal outcomes, with epigenetic dysregulation increasingly implicated in fetal programming. As the placenta plays a central role in maternal–fetal nutrient and hormonal exchange, this study investigated structural, epigenetic, and metabolic alterations in placentas from GDM pregnancies. Pregnant women recruited from PRS Hospital Pvt. Ltd. were classified as normoglycemic and GDM based on oral glucose tolerance test results, and maternal blood, cord blood, and placental tissues were collected for histological evaluation, DNA methylation profiling, lipid analysis, and gene expression studies. GDM placentas exhibited pronounced morphological abnormalities, including increased syncytial knots, chorangiosis, villous immaturity, and fibrinoid necrosis. Epigenomic analysis identified 521 differentially methylated regions, with significant enrichment in lipid metabolism pathways; notably, the lipid-associated gene PLIN1 was hypermethylated and significantly downregulated in GDM placentas. Concordantly, lipid profiling of maternal and cord blood revealed marked dyslipidemia in GDM subjects, indicating disrupted lipid homeostasis at both molecular and systemic levels. Collectively, these findings demonstrate that GDM induces coordinated structural, epigenetic, and metabolic alterations in the placenta that may influence fetal lipid exposure and contribute to long-term metabolic risk, highlighting PLIN1 as a potential biomarker and therapeutic target.

RNA sequencing data of whole blood cells of normal glucose tolerant (NGT) and gestational diabetes (GDM) pregnant women

Project description:Total RNA was isolated from WBCs. For the analysis of genome-wide expression differences in small non-coding RNAs (sncRNAs) and long-coding and non-coding RNAs (mRNAs and lncRNAs), NGT and GDM pregnant women were selected. Twenty-nine GDM-associated mature micro-RNAs (miRNAs) with increased expression and one hundred sixty-three mRNAs with reduced expression associated with GDM were found (P<0.05 and FDR<0.1).

RNA sequence of gestational diabetes mellitus (GDM) and healthy control placentas

Project description:GDM is a multi-system disorder that is primarily characterised by new-onset hypertension accompanied by proteinuria during gestation. This disease is one of the leading causes of maternal and perinatal morbidity and mortality. In this work, placental samples were collected from GDM and control patients. RNA-seq was performed to identify differences in gene expression. Significantly differentially expressed genes between the GDM and control samples included 64 up-regulated and 296 down-regulated genes. This study may provide further insight in the mechanisms of GDM and function as preventive, predictive and therapeutic measures.

Expression profiles of placenta and umbilical cord blood with or without gestational diabetes mellitus (GDM)

Project description:Objective: To explore the characteristics and underlying molecular mechanisms of genome-scale expression profiles of women with- or without- GDM and their offspring. Materials and Methods: We recruited a group of 21 pregnant women with GDM and 20 healthy pregnant women as controls. For each pregnant women, RNA-seq were performed using the placenta and paired neonatal umbilical cord blood specimens. Differentially expressed genes (DEGs) were identified with BMI of pregnant women as covariates. Then, functional enrichment analysis was performed separately or interactively in placenta and umbilical cord blood. Results: Through the comparison of GDM and healthy samples, 1442 and 488 DEGs were identified from placenta and umbilical cord blood, respectively. Functional enrichment analysis showed that the placenta expression profiles of GDM women mirrored the molecular characteristics of type II diabetes and insulin resistance patients. DEGs illustrated significant overlaps among placenta and umbilical cord blood samples, and the overlapping DEGs were associated with endocrine resistance and insulin resistance. Conclusions: Our research demonstrated the transcriptomic alternations of GDM mothers and offspring. Our findings emphasized the importance of epigenetic modifications in the communication between pregnant women with GDM and offspring, and provided reference for the prevention, control, treatment, and intervention of perinatal deleterious events of GDM and neonatal complications.

Project description:Gestational diabetes (GDM) refers to diabetes diagnosed as diabetes in the second or third trimester of pregnancy, but not obvious before pregnancy. It is one of the most common complications of pregnancy, which can lead to perinatal asphyxia, neonatal hypoglycemia, macrosomia, cardiovascular abnormalities, obesity and other short-term and long-term adverse outcomes of the offspring. This study explores the differential expression of transcriptome between GDM and non GDM placenta, and explores the impact of subtle metabolic changes in the placenta on fetal growth and development during GDM in pregnant women.

Circulating lncRNAs NONHSAT054669.2 and ENST00000525337 can be used as early biomarkers of gestational diabetes mellitus

Project description:This study aimed to identify key circulating long non-coding RNAs (lncRNAs) as novel biomarkers for diagnosis of GDM at the early stages. First, lncRNA microarray analysis was conducted for plasma samples of GDM women before delivery and 48 h after delivery. The expression of differentially expressed lncRNAs in clinical samples at different trimesters was randomly validated by quantitative PCR. Moreover, the correlation between lncRNA expression and oral glucose tolerance test (OGTT) level in GDM women during the second trimester was analyzed, and the diagnostic value of key lncRNAs for GDM during different trimesters was evaluated by receiver operating characteristic (ROC) curve. The plasma level of NONHSAT054669.2 and ENST00000525337 may be used as novel diagnostic biomarkers for early diagnosis of GDM.

High-throughput sequencing analysis of placental tissue from pregnant women with gestational diabetes and gestational hypertension

Project description:In this study, transcriptome sequencing was conducted on placentas obtained from both normal pregnancies and pregnancies with GDM to investigate the molecular mechanisms underlying this condition. Placental tissue samples were immersed in ice-cold phosphate-buffered saline (PBS) to remove blood contamination. Sections with a thickness of 4 µm were cut from the block and stained with Hematoxylin and eosin (HE) staining. The resulting slides were examined and photographed under a light microscope (Olympus).The original sequencing data were subjected to sequencing analysis, resulting in the identification of 935 upregulated genes and 256 downregulated genes. The Kyoto Encyclopedia of Genes and Genomes and Gene Ontology analysis techniques on differential genes uncovered evidence suggesting that the phosphoinositide 3-kinase (PI3K) /Akt signaling pathway may contribute to the pathogenesis of GDM. Subsequent analysis indicated that the expression levels of MMP11, MMP12, MMP14, and MMP15, which are regulated by the PI3K/Akt pathway, were upregulated in the placentas of patients with GDM when compared to those of individuals with normal placental function. Additionally, our investigation into alternative splicing patterns revealed an increase in exon skipping alternative splicing of CSF3R in the placenta of patients with GDM compared to that in the control group.
