ABSTRACT: To explore the impact of nasal commensal viruses on the onset and progression of allergic rhinitis, We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of CD45+ cells in the nasal mucosa of control and AR mice treated with Vehicle or Ribavirin.

We used microarrays to screen gene expression profiles in nasal mucosal epithelial cells of allergic rhinitis, and predicted the relationship between lncRNA and mRNA by bioinformatics analysis.

Exon Array Analysis of Asthmatic Chronic Rhinosinusitis with Nasal Polyps

Project description:Asthmatic chronic rhinosinusitis with nasal polyps (aCRSwNP) is a common disruptive eosinophilic disease. Therefore, we sought to identify gene expression changes in nasosinus inflamed mucosa and adjacent polyp tissue from subjects with aCRSwNP. Twelve asthmatic chronic rhinosinusitis (aCRS) subjects (11 with nasal polyps; aCRSwNP) provided inflamed nasosinus mucosa (11 samples) and adjacent polyp (10 samples) or a small mixed mucosa and polyp specimen (1 sample) (thus, 22 samples overall). Inflamed mucosa or polyp was from the middle meatus or anterior ethmoid cavity. As control samples, nasosinus tissue was collected from the inferior turbinate or uncinate process from normal or rhinitis subjects without nasal polyps (n=17); 4 had physician-diagnosed allergic rhinitis (AR), 2 had suspected AR, 1 had suspected vasomotor rhinitis, and 10 had no signs of nasosinus disease, allergy, or atopy. All 4 AR subjects chose to donate tissue outside of their known allergy season(s).

Project description:To elucidate the potential role of commensal viruses in the etiology of AR, we performed a metagenomic analysis of nasal mucosa to identify commensal viruses in the nose of mice. To evaluate the potential role of these commensal viruses in AR, we reduced them in the nose of mice by administrating intranasal drops of vehicle (Vehicle group) or broad-spectrum antiviral drug ribavirin daily, starting at 4-week-old and lasting for three weeks (Ribavirin group). These mice were also administrated with PBS, OVA, or HDM to induce AR.
