ABSTRACT: Human naïve CD4 T cells were purified from healthy volunteers' peripheral blood using commercially available kits and differentiated into effector Th1 in vitro by culturing for 6 days in the presence of anti-CD3 and CD28 antibodies (2 µg/ml each) and IL-12 (20 ng/ml). Human effector Th1 cells were harvested on day 6 post-differentiation and treated with either vehicle, LY (0.0125 µM), or Methyl Pyruvate (MePyr, 5 mM) in complete RPMI media for 24 hours.
