L1CAM promotes human endometrial cancer via NF-κB activation

ABSTRACT: RNA-seq analysis of human endometrial carcinoma HHUA cells was performed to determine the impact of L1CAM knockdown. Cells were transduced with shRNA targeting L1CAM or shscramble and harvested 48 hours later for total RNA extraction. This dataset provides insight into gene expression changes associated with L1CAM suppression in uterine cancer.

Development of antibody-drug conjugates targeting L1CAM to treat metastatic cancer

Project description:Effective treatments for metastatic disease have eluded precision oncology efforts and represent a major unmet need. The L1 cell adhesion molecule (L1CAM) is a transmembrane protein expressed by populations of Metastasis Stem cells (MetSC), that are enriched for the ability to reinitiate and propel metastatic growth and display phenotypic plasticity and resistance to conventional chemotherapies. Notably, L1CAM+ MetSC have been detected in numerous human solid tumor types and in multiple disseminated organ sites. As a selective marker of MetSC, L1CAM is a promising candidate for molecularly targeted drugs aimed at eliminating metastases, yet strategies to date have not achieved clinical success. Here, we develop antibody-drug conjugates to deliver highly toxic PNU-159682 payloads to L1CAM-expressing cells. We report the generation of monoclonal antibodies (mAb) with high binding affinity, specificity and selectivity for the human L1CAM extracellular domain. After further optimization of the L1CAM mAbs, PNU-159682 was conjugated to generate ADC variants with both cleavable and non-cleavable linkers, with an average drug-antibody-ratio (DAR) of four. The three ADCs (Y-004, Y-005 and Y-006) potently killed cells with varying levels of surface L1CAM expression. L1CAM ADCs given as monotherapy resulted in robust tumor growth control and extended survival in mice harboring subcutaneous L1CAM+ xenografts or L1CAM+ lung metastases, with safety analyses indicating a feasible therapeutic window. Our findings provide strong proof-of-concept to extend preclinical development of these novel L1CAM ADCs as therapeutic agents for advanced cancer.

Analysis of L1CAM-regulated signaling pathways and cellular responses to anti-L1CAM radioimmunotherapy in human ovarian cancer cells

Project description:L1 cell adhesion molecule (L1CAM) is overexpressed in various solid tumors, and its overexpression is linked to increased invasion, metastasis, angiogenesis, cancer stemness, and therapy resistance. However, the mechanisms of L1CAM-mediated carcinogenesis and radioresistance, particularly in high-grade serous ovarian carcinoma (HGSOC), the most common and lethal type of ovarian cancer, remain insufficiently understood. Anti-L1CAM radioimmunotherapy (RIT) using the radioisotopes Terbium-161 (Tb-161) and Lutetium-177 (Lu-177) has shown that Tb-161 is more cytotoxic than Lu-177, due to its distinct radioactive properties. Moreover, we identified a population of L1CAM+/CD133+ cells as cancer stem cells (CSCs) in ovarian cancer and demonstrated L1CAM's association with radioresistance. Understanding L1CAM-regulated downstream signaling and cellular responses to anti-L1CAM RIT with Tb-161, compared to Lu-177, is now critical. To address this, we established CRISPR-Cas9-mediated L1CAM knock-out (ΔL1CAM) in human ovarian cancer cells (OVCAR8) alongside a control cell line with a knockout of the nonessential gene AAVS1 (ΔAAVS1). In this study, quantitative phosphoproteomics, coupled with matching proteomics, revealed (i) L1CAM-dependent signaling pathways and biological processes by comparing ΔL1CAM with wild-type cells, and (ii) cellular responses to anti-L1CAM RIT with Tb-161 and Lu-177 in L1CAM-expressing ovarian cancer cells.

single cell mRNA sequencing of B cells from human endometrial cancers

Project description:Emerging data indicate that B-cells play a key role in cancer suppression, particularly when aggregated in tertiary lymphoid-like structures (TLS). Here, we used single-cell and bulk RNA sequencing of endometrial cancers (ECs) and found L1-cell adhesion molecule L1CAM to be significantly associated with TLS. Immunohistochemical (IHC) expression of L1CAM was restricted to the germinal centre (GC) of mature TLS, where it co-localised with follicular dendritic cell (FDC) marker CD21. Tumours surrounded by L1CAM-positive TLS were identified across all molecular EC subgroups, but enriched in mismatch repair deficient (MMRd) and polymerase-epsilon mutant (POLEmut) EC, especially when TP53 mutations were also present. Importantly, L1CAM-positive TLS appeared an independent favourable predictor of EC recurrence and death, and significantly improved the current prognostic molecular EC classification based on clinicopathological and molecular factors. Our data suggest a pivotal role of TLS in predicting outcome of EC patients, and establishes L1CAM as a simple and widely available biomarker of TLS in clinical care.

The effect of obesity on uterine receptivity is mediated by endometrial extracellular vesicles that control human endometrial stromal cell decidualization and trophoblast invasion

Project description:The objectives of the present study were to determine whether obesity impacts human decidualization and the endometrial control of trophoblast invasion (both of which are required for embryo implantation) and evaluate the potential involvement of endometrial extracellular vesicles (EVs) in the regulation of these physiological processes. Using primary human cell cultures, we first demonstrated that obesity is associated with significantly lower in vitro decidualization of endometrial stromal cells (ESCs). We then showed that the trophoblastic cell line’s invasive ability was greater in the presence of conditioned media from cultures of ESCs from obese women. Using mass-spectrometry-based quantitative proteomics, we found that EVs isolated from uterine supernatants of biopsies from obese women (vs. nonobese women) presented a molecular signature focused on cell remodeling and angiogenesis. Lastly, the results of functional assays indicated that supplementation of the culture medium with EVs from nonobese women can rescue (at least in part) the defect in in vitro decidualization described in ESCs from obese women. Lastly, the addition of endometrial EVs from obese women (vs. nonobese women) was associated with significantly greater invasive activity by HTR-8/SVneo cells. In conclusion, our results provided new insights into the endometrial EVs’ pivotal role in the poor uterine receptivity observed in obese women.

Endometrial gland impacts on stromal cell decidualization

Project description:Uterine glands and, by inference, their secretions impact uterine receptivity, blastocyst implantation, stromal cell decidualization, and placental development. Changes in gland function across the menstrual cycle are impacted by steroid hormones, estrogen and progesterone, as well as stroma-derived factors. Using an endometrial epithelial organoid (EEO) system, transcriptome and proteome analyses identified distinct responses of the EEO to steroid hormones and prostaglandin E2 (PGE2). Notably, steroid hormones and PGE2 modulated the basolateral secretion of EEO proteins, where cystatin C (CST3) was significantly increased by progesterone and PGE2. CST3 treatment of decidualizing stromal cells significantly decreased the decidualization markers PRL and IGFBP1. The attenuation of stromal cell decidualization via CST3 suggests a role for uterine gland-derived proteins in controlling the extent of decidualization. These findings provide evidence that uterine gland-derived factors directly impact stromal cell decidualization, which has strong implications for better understanding pregnancy establishment and female fertility in humans.
