Single cell gene expression profiling of triple negative breast cancer organoids

ABSTRACT: Triple negative breast cancer (TNBC) is a heterogenous disease driven by aberrant activation of receptor tyrosine kinases and the proliferative MEK-ERK signaling pathway. Heterogeneity in kinase expression levels across two TNBC organoid lines was used in combination with the Atlas of substrate specificities for the human Ser/Thr kinome to predict phenotypic differences between the two organoid lines. Single nucleus RNAseq validated these predictions showing high enrichment of immune and inflammatory signatures within a subpopulation of tumor cells.
