Runx1 is a mediator of fibrotic activation and fibrotic memory in lung fibroblasts

ABSTRACT: Runx1 emerged as a transcription factor that was potentially regulating fibroblast activation and memory following intratracheal bleomycin injury. In order to examine this, fibroblasts were lineage traced and collected following bleomyicn induced fibrosis at the peak fibrotic time point, Day 14, and at the time of fibrotic recovery, Day 56. Runx1 ChIPseq was performed to assess Runx1 binding in regions of genes known to be associated with fibrosis.

Transcriptional profile of mouse lung during the fibrotic phase of the response to bleomycin

Project description:Wildtype mice were given saline or bleomycin by oropharyngeal instillation. After 14 days, during the fibrotic phase of the response, lungs were dissected and total RNA was extracted and used for gene expression profiling. The aim was to identify those genes regulated during the development of fibrosis in this animal model of bleomycin-induced lung fibrosis. Wildtype male C57Bl/6J mice (8-10 weeks old) were used in the study, with three mice per group. Bleomycin (1mg/kg body weight in 50μl of saline) or saline was administered by oropharyngeal installation as described previously by Lakatos et al, Exp Cell Res, 2006, under light halothane-induced anaesthesia. After 14 days, lungs were removed, blotted dry and the trachea and major airways were excised before the separated lobes were snap frozen in liquid nitrogen. Lungs were then pulverized under liquid nitrogen, and the resulting lung powder was used for total RNA extraction using Trizol. Following DNase-treatment, clean-up, cDNA synthesis and cRNA synthesis, samples were hybridized to Affymetrix MOE430A genechips using standard protocols. The data were analyzed using RMA with quantiles normalization.

Project description:To identify the underlying mechanisms associated with lung and kidney fibrosis inducing lung metastasis, we evaluated global transcriptomic changes in healthy lung, fibrotic lung fibrosis, and lung from mice with kidney fibrosis. Results provide information on the vascular remodeling of the lungs with organ fibrosis. Lung fibrosis is induced with intratracheal bleomycin administration. Kidney fibrosis is induced by unilateral ureter obstruction (UUO).

Bulk RNA sequencing analysis of human airway basal stem cells stimulated by BALF from mouse fibrotic lungs and normal lungs

Project description:To understand how the microenvironment of fibrotic lungs affects the behavior of airway basal stem cells (BCs), we constructed a mouse model of pulmonary fibrosis using bleomycin induced injury, and collected bronchoalveolar lavage fluid (BALF) from the control group and bleomycin damaged mice on day 14 after injury. Then, human BCs were stimulated with control or bleomycin BALF in culture and subjected to bulk RNA sequencing. Transcriptomic analysis showed that basal stem cells stimulated with bleomycin BALF upregulated genes related to chemotaxis and migration, and activated the TGF-β pathway.

Project description:Idiopathic pulmonary fibrosis (IPF) is a progressing chronic and fibrotic lung response with poor prognosis. To study the underlying molecular mechanisms of IPF, the rat bleomycin model is commonly used. Intratracheal application of bleomycin is known to induce inflammatory and fibrotic processes in the lung, e.g. infiltratin of inflammatory cells and collagen deposition. Recently, the PDE4B inhibitor BI 1015550 has been reported to prevent a decrease in lung function in patients with IPF. Here we examined the effect of PDE4 inhibitor BI 1015550 in the rat bleomycin model on transcriptional level by RNASeq.

Sustained Yap/Taz activation promotes aberrant alveolar epithelial cell differentiation and drives persistent fibrotic remodeling

Project description:YAP/TAZ signaling is required for initiation of lung alveolar repair, yet previous studies in idiopathic pulmonary fibrosis (IPF) predicted increased YAP/TAZ signaling in alveolar epithelial cells (AECs). We investigated whether persistent YAP/TAZ AEC signaling contributes to failed epithelial repair and persistent fibrotic remodeling. In IPF lungs, we identified increased YAP+/TAZ+ AECs and increased transcriptional target expression. Pharmacological YAP/TAZ activation in human AEC organoids and in murine AT2 cell organoids generated with genetic Yap/Taz activation (YTactive) (via deletion of Hippo-kinases Stk3/4), resulted in phenotype shifts into aberrant transitional and airway-like states. Bleomycin injury of YTactive mice resulted in persistent fibrotic remodeling at 28- and 56-days post-bleomycin injury. Gene promoter activity associated with transitional cell markers (Krt19, Hopx, and Runx2) was increased in YTactive AT2 cells. Immunofluorescent staining showed a loss of AT2 associated Cebpa and increased Krt19 in YTactive lineage traced AT2 cells 28 days post-injury. Inhibition of Yap/Taz using Verteporfin resulted in improved lung repair in YTactive mouse lungs, including restored Cebpa and decreased Krt19+ transitional cells. These findings demonstrate sustained Yap/Taz activation drives abnormal alveolar repair and persistent fibrotic remodeling. Blocking aberrant persistent Yap/Taz activity promotes adaptive repair and has potential as a therapeutic strategy for pulmonary fibrosis.

HSP27 (Heat Shock Protein 27, HSPB1) overexpression effect on bleomycin-induced PF (Pulmonary fibrosis) mice model

Project description:To analyze the role of HSP27 in bleomycin (BLM)-induced pulmonary fibrosis in vivo, we used TG mice that conventionally overexpressed HSP27. Our previously reported that HSP27 plays an important role in pulmonary fibrosis. In mice overexpressing HSPB1, BLM-induced fibrotic regions and collagen deposits were increased compared with those in control BL6 mice. Following intratracheal treatment of BLM, the HSP25 TG mice showed an abundance of mononuclear and neutrophils cells in the alveoli, greater destruction of alveolar septa, intra-alveolar hyaline membrane formation, and a marked increase in collagen deposition compared with control BL6 mice.
