Project description:Tire wear particles (TWP) are small micro- or nano-particles resulting from the friction of tire tread against roads. These microplastics have been found in waterways, usually through rain and wind, posing a potential risk to exposed aquatic life. The goal of this project was to assess the toxicity of chemical leachates from TWP and characterize how they may impact the health and development of aquatic life, like Danio rerio, zebrafish. Furthermore, we also determined if solar-simulated or dark conditions intensified the effects of TWP, considering processes like photodegradation and photochemical oxidation occur in sunlight. Zebrafish were exposed from 0-4 days post fertilization (dpf) to a filtered TWP solution at varying concentrations, and each concentration was leached in either solar-simulated or dark conditions. Mortality and hatching rates were quantified throughout the exposures, and physical anomalies were recorded at 4 dpf using microscopy. Ethyoxyresorufin-O-deethylase (EROD) activity, an indicator of Phase I biotransformation, was performed using fluorescent microscopt. RNA sequencing was performed for 4 dpf larvae. TWP exposures, regardless of leaching light status, increased mortality, yolk sac edema, pericardial edema, spinal curvature, and craniofacial malformations, while decreasing hatching and swim bladder inflation. Regardless of lighting during leaching, EROD activity increased in the liver and brain and decreased in the gut CYP1A. RNA sequencing revealed some conserved processes in response to TWP, regardless of light status during leaching, including those involved in neurobiological function. However, divergent responses were also observed for processes such as amino acid metabolism, steroid biosynthesis, and fatty acid metabolism. Overall, these results suggest a disruption of embryonic development in zebrafish when exposed to TWP, and that sunlight during chemical leaching can alter the molecular responses to TWP.

Project description:Road dust cotains tire wear particles (TWPs) and a large amount of mineral particles (MPs). Given that tire tread in vehicles is mainly comprised of natural rubber (NR), isoprene and dipentene could be the main pyrogenic products stemmed from the thermolysis of NR. This offers a great chance to quantify the exact mass of TWP in road dust. As such, this study focused on the influence of MPs on the trends in thermolytic behaviors of NR using the resistive furnace (furnance) and Curie point pyrolyzers. This study confirmed that a reliable correlation in line with the formation of isoprene and dipentene could not be realized using the furnace type of a pyrolyzer. This means that employing the furnace type of a pyrolyzer in quantitification of TWPs could not be a viable and approproiate option due to the diverted thermolytic trends of NR due to differences in the heat transfer and adsoprtion of the pyrogenic products triggered by MPs. In the Curie point type of a pyrolyzer, the production rates of isoprene and dipentene were linearly responded to the mass of NR. The ferromagnetic substance in MPs could lead to the thermolytic trend change of NR. Thus, adopting the Curie point type of a pyrolyzer could be a viable option for quantification of TWPs in road dust when the effects of ferromagnetic substance are well neutralized.

Project description:The goal of the study was to compare gene expression of Robo1+/+ and Robo1-/- luminal progenitors. Total RNAs were then extracted from FACS purified luminal progenitor cells, harvested from Robo1+/+ or Robo1-/- mice (n=3 per genotype, two animals per n) using TRIreagent LS (Sigma, T3934). Poly(A)+ RNA sequencing libraries were made from each sample using the TruSeq RNA library preparation kit v.1 (Illumina). Illumina RNA PolyA library preparation guide. A total of 6 libraries were created by PCR amplification with Illumina barcoding primers using kit recommended conditions and quantified using a Bioanalyzer DNA 1000 kit (Agilent).

Project description:Tire wear particle (TWP)-derived compounds may be of high concern to consumers when released in the root zone of edible plants. We exposed lettuce plants to the TWP-derived compounds diphenylguanidine (DPG), hexamethoxymethylmelamine (HMMM), benzothiazole (BTZ), N-phenyl-N'-(1,3-dimethylbutyl)-p-phenylenediamine (6PPD), and its quinone transformation product (6PPD-q) at concentrations of 1 mg L-1 in hydroponic solutions over 14 days to analyze if they are taken up and metabolized by the plants. Assuming that TWP may be a long-term source of TWP-derived compounds to plants, we further investigated the effect of leaching from TWP on the concentration of leachate compounds in lettuce leaves by adding constantly leaching TWP to the hydroponic solutions. Concentrations in leaves, roots, and nutrient solution were quantified by triple quadrupole mass spectrometry, and metabolites in the leaves were identified by Orbitrap high resolution mass spectrometry. This study demonstrates that TWP-derived compounds are readily taken up by lettuce with measured maximum leaf concentrations between ∼0.75 (6PPD) and 20 μg g-1 (HMMM). Although these compounds were metabolized in the plant, we identified several transformation products, most of which proved to be more stable in the lettuce leaves than the parent compounds. Furthermore, continuous leaching from TWP led to a resupply and replenishment of the metabolized compounds in the lettuce leaves. The stability of metabolized TWP-derived compounds with largely unknown toxicities is particularly concerning and is an important new aspect for the impact assessment of TWP in the environment.

Project description:The ecological impact of tire wear particles in aquatic ecosystems is a growing environmental concern. We combined toxicity testing, using fathead minnow (Pimephales promelas) embryos, with nontarget high-resolution liquid chromatography Orbitrap mass spectrometry to characterize the toxicity and chemical mixture of organic chemicals associated with tire particle leachates. We assessed: 1) exposure to tire particle leachates after leaching for 1-, 3-, and 10-d; and 2) the effect of the presence and absence of small tire particulates in the leachates. We observed a decrease in embryonic heart rates, hatching success, and lengths, as well as an increase in the number of embryos with severe deformities and diminished eye and body pigmentation, after exposure to the leachates. Overall, there was a pattern whereby we observed more toxicity in the 10-d leachates, and greater toxicity in unfiltered leachates. Redundancy analysis showed that several benzothiazoles and aryl-amines were correlated with the toxic effects observed in the embryos. These included benzothiazole, 2-aminobenzothiazole, 2-mercaptobenzothiazole, N,N'-diphenylguanidine, and N,N'-diphenylurea. However, many other chemicals characterized as unknowns are likely to also play a key role in the adverse effects observed. Our study provides insight into the types of chemicals likely to be important toxicological drivers in tire leachates, and improves our understanding of the ecotoxicological impacts of tire wear particles. Project description:Particulate matter (PM2.5) samples were collected at two different places of a college campus (CC) and a bus stop (BS) nearby the college campus. The traffic volume of college campus was very low due to untact classes. Polymeric components and tire wear particle (TWP) contents in the PM2.5 samples were analyzed using pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS). Various polymeric components such as natural rubber (NR), bitumen, saturated hydrocarbons, poly(ethylene terephthalate) (PET), and plant-related particles (PRPs) were observed. NR and bitumen are key components of TWP of bus tire tread and asphalt pavement wear particle (APWP), respectively. The TWP contents in the PM2.5 samples collected at the bus stop were larger than those collected at the college campus. For the same sampling site, the TWP content in the PM2.5 sample collected for higher fine dust concentration in the air was greater than that for lower one. The TWP2.5 concentration in the air for the BS sampling was higher than those for the CC sampling, even when the PM2.5 concentration in the air for the former was lower than those for the latter. It can be concluded that the TWPs and APWPs in the PM2.5 samples collected at the college campus should be transferred mostly from the outside road.

Project description:Acute kidney injury (AKI) is an important contributor to the development of chronic kidney disease (CKD). We performed miRNA and mRNA sequencing on biobanked human kidney tissues obtained in the routine clinical care of patients with the diagnoses of AKI and minimal change disease (MCD), in addition to nephrectomized (Ref) tissue from individuals without known kidney disease. From all renal biopsy samples, 2 cryosections (10 μM) including the entire cross-section of the tissue were placed directly into PicoPure RNA extraction buffer. RNA was isolated using the PicoPure isolation kit. Total RNA was evaluated for quantity and quality using an Agilent Bioanalyzer. Approximately 10 ng of total RNA were used for each sample’s library preparation. Ribosomal RNA was removed using RiboGone – Mammalian Kit protocol. After rRNA depletion, double-stranded cDNA was synthesized and amplified using the SMARTer Universal Low Input RNA Kit protocol. The cDNA was sheared by Covaris, and the library was prepared and barcoded following the Ion Plus Fragment Library Kit protocol. Each resulting barcoded library was quantified and quality assessed by Agilent Bioanalyzer. Multiple libraries were pooled in equal molarity. Finally, 8 μL of 100 pM pooled libraries were loaded into lanes of an Illumina HiSeq 4000. At least 30M reads per library were generated.
