RNA-seq and ATAC-seq of Psc Su(Z)2 -/- intestinal stem cells in Drosophila

ABSTRACT: Polycomb group (PcG) genes function as epigenetic silencers that maintain the repressive states of target genes essential for normal development. To elucidate the role of PRC1 components in the Drosophila midgut, we performed both RNA-seq and ATAC-seq analyses on intestinal stem cells (ISCs) deficient for PRC1 and Su(z)2. For RNA-seq controls, besides the esgts>GFP, we included ISCs harboring mutations in the Notch signaling pathway (N55e11) and the JAK-STAT pathway (dome468) as additional controls. What's more, ATAC-seq was also performed on pc-RNAi and E(z)-RNAi ISCs which represent other components of PRC1 and PRC2, respectively.
