Characterization of miRNA expression in 3D cultured cells

ABSTRACT: A Comprehensive miRNA-lncRNA-mRNA Corregulation Network in 3D Cultures of Luminal B Breast Cancer Three-dimensional (3D) cell cultures offer advantages over traditional 2D models by better replicating tumor biology. In this study, we examined the effects of 3D culture on miRNA expression in BT-474 luminal B breast cancer cells. Microarray analysis revealed significant differences: 82 miRNAs were upregulated and 119 downregulated in 3D conditions. A regulatory network was constructed involving four miRNAs consistently downregulated (miR‑92a‑3p, miR‑539‑5p, miR‑18a‑5p, miR‑130a‑3p), potentially influencing MAPK signaling, ECM remodeling, and estrogen receptor activity. These results highlight how 3D systems improve understanding of breast cancer biology and may help uncover novel therapeutic targets.
