Endothelial miR-33 protects from atherosclerotic lesion initiation and acute inflammation


ABSTRACT: Human aortic endothelial cells (HAEC) were treated with TNF, or TNF and LDL, in order to determine how cholesterol loading influences HAEC inflammatory responses to TNF. Cells were also transfected with a miR-33 mimic to determine which of the LDL-mediated perturbations are dependent on cholesterol-induced miR-33 dysregulation. From these analyses, we determined that miR-33 is able to rescue a signifcant portion of the genes dysregulated in hypercholesteremic inflammation compared to normocholesteremic inflammation.
